Mouse are commonly used in experimental animals.According to the previous literature on hair follicle formation and development,combined with the experimental can be seen hair follicle began to form in the embryonic period 13th days of the mouse,hair follicle formation in embryonic period 17th days of the mouse.After birth,hair follicle anagen period is 1th to 11th days,hair follicle catagen period is 12th to 18th days,hair follicle telogen period is 19th and 20th days,hair follicle from 21th days began into the next growth cycle.Fibroblast growth factor 7(FGF7)subfamilies(including FGF7,10 and 22)play an important role in skin development and repair.FGF7 play an important role in initiate the new hair cycle.FGF10 as a promising agent for development of agents for human hair loss treatments.FGF 22 plays an important role in skin development and repair.The effect of the FGF7 subfamily on hair formation and development was not reported in the stage where the hair follicle began to form to the end of the first follicle cycle.In order to study the effect of FGF7 subfamily on the formation and growth of hair follicles in this stage,this study by real-time quantitative PCR,Western blot and immunohistochemistry to research the distribution of FGF7,10 and 22 in mouse during the hair follicle began to form to the first hair follicular cycle.To show that FGF7,10,22 gene and protein differential expression of mouse skin during hair follicles began to form to the first hair follicle cycle,to explore the effects of its subfamily on hair follicle formation and development,to further reveal the mechanism of hair growth adjustment to lay the foundation.The results are as follows:(1)The results of immunohistochemistry showed that FGF7 was mainly expressed in the hair germs cell and epidermis during hair follicles began to form period,and was mainly expressed in the innner root follicle and epidermis of the hair follicle in the first hair follicle cycle.The RT-PCR and Western blot showed that the expression level of FGF7 on the 13th day of hair follicle formation was the lowest at the 14th day in period of hair follicles began to form,and the expression level of FGF7 decreased gradually with the increase of the number of days.The FGF7 was the lowest in hair follicle catagen during the first hair follicle cycle and was highly expressed at telogen and anagen periods.(2)The results of immunohistochemistry showed that FGF10 was mainly expressed in the hair germs cell and epidermis during hair follicles began to form period,and was mainly expressed in the inner/outer root follicle,Sebaceous glands and epidermis in the first hair follicle cycle.The RT-PCR and Western blot showed that the expression level of FGF10 on the 13th day of hair follicle formation was the lowest at the 15th day in period of hair follicles began to form,and the expression level of FGF10 decreased gradually with the increase of the number of days.The FGF10 was the lowest in hair follicle catagen during the first hair follicle cycle and was highly expressed at telogen and anagen periods.(3)The results of Immunohistochemical showed that FGF22 was mainly expressed in the Bulbous hair peg and epidermis during hair follicles began to form period,and was mainly expressed in the outer root sheath,epidermis and sebaceous glands during the first hair follicle cycle.The RT-PCR and Western blot showed that the expression level of FGF22 on the 14th day was the lowest,and then the expression level of FGF22 increased gradually in period of hair follicles began to form.The expression level of FGF22 hair follicle was the highest in the catagen of the first hair follicle cycle,and was low at telogen and anagen periodsIn the period of hair follicles began to form,these results suggest that FGF7,10 participates in instructing hair germ cells to proliferate,and may play an important role in the early stage of hair follicle formation.FGF22 plays an important role in hair follicle formation and hair follicle maturation.FGF7,10 plays an important role in the induce hair follicle into the new cycle during the development of hair follicle,and FGF22 has an important effect to induce hair follicle into the catagen period.
