During the period of the first egg laying,the physiology of laying hen undergoes the dramatically changes and Feed Conversion Ratio(FCR)is improved for laying eggs.The objective of this study was to analyze the changes of the liver metabolic function and the effects of the changes on nutrients conversion and utility based on differentially expressed gene analysis in the intestine(duodenum?jejnum and ileum)and the liver.One week before first laying,before laying,the day of first laying and one week after first laying,full-sib White Leghorn layers were chosen,hormone(GH?INS and PRL)?the length and weight of intestine in four physiological periods were measured,liver and intestine sampling the transcriptome data were obtained by using RNA-seq and the gene expression differences was compared before and after the first egg laying.By analysis the obtained date and sequencing,there is no differently changed between hormone and the length and weight of intestine(P>0.05);105,148 and 109 differentially expressed genes were isolated from the duodenum,jejunum and ileum according to the intestinal transcriptome in two physiological periods one week before and after first egg laying.GO functional annotation and KEGG metabolic pathway analysis of differentially expressed genes showed that these genes mainly involved in cell membrane synthesis,tissue development and stress response.Two hundred and twenty-two differently expressed genes which have more than 2-fold changed were obtained in liver transcriptome(P<0.05),and some genes mainly involved in lipid metabolism and tryptophan metabolism.In this study,there have some function-unknown genes were found,such as ring finger protein 186(RNF186).We obtained the liver differently expressed genes from White Leghorn birds before and after first eggs laying and analyzed the changes of metabolism pathway of nutrients in the liver.These results proved the liver metabolism pathway of nutrients were changed by these differently expressed genes and the changes after first egg laying could improve the nutrients utilization of the feed.
