The TaEF and TaCP wheat expression genes were induced by low phosphorus stress,which screened by c DNA-AFLP technology early time in our lab,it may play an important role in response mechanism to low phosphorus stress of wheat.The biological functions of the two genes were analyzed and studied in this paper.(1)TaEF expression analysis in different wheat varieties under the condition of phosphorus stress: the material were wheat Jimai17(JM17),Zhengmai 9023(ZM9023),Xinong 979(XN979),Xiaoyan 54(XY54)and Yunong 202(YN202),the ground and underground part of wheat were respectively took at 7 days,14 days,21 days,phosphorus in 1 days,normal phosphorus and phosphorus deficiency,and the fluorescent quantitative PCR technology was used to express the quantity analysis on5 different wheat varieties.The results showed that the TaEF gene raised the expression at root and leaf in ZM9023,XY54 and YN202,and decreased in XN979 under the condition of phosphorus deficiency.It is showed that the TaEF gene may be involved in the related gene transcription regulatory mechanism on wheat varieties under the low phosphorus stress,but the performance is different in different wheat varieties.(2)TaEF functional analysis in transgenic Arabidopsis thaliana: Transfer the c DNA of TaEF gene into the plant expression,and into the Arabidopsis thaliana by the method of agrobacterium-mediated,and the over expression plants were built.It is showed that the transgenic plants were successfully built after the test analysis of the transcription and protein expression level.Physiological expression of Arabidopsis lines indicated that over expression of TaEF in Arabidopsis to produce significant changes in phosphorus stress,showing the growth and development characteristics of low phosphorus adaptability.Overexpression of five different strains and wild type Arabidopsis respectively were cultured in two cases of MS culture medium and water culture,the differences of physiological changes in phenotype were observed between over expression strains and wild type.The root length and plant height of wild type were less than that of over expression strains in the absence of phosphorus,low phosphorus(5?M)and normal phosphorus circumstances when cultured in the medium of MS 14 days;and the ground part weight of the wild type was lower than that of over expression strains at phosphorus deficiency and normal phosphorus,but higher at low phosphorus(5?M).The root length of wild type were less than that of over expression strains in the absence of phosphorus,low phosphorus(5?M)and normal phosphorus circumstances when cultured in water,but the root weight was above the over expression type in phosphorus deficiency,and the ground part weight of the wild type was less than that of over expression strains at phosphorus deficiency and normal phosphorus.(3)TaCP gene expression analysis.q PCR analysis results showed that the TaCP gene was expressed increasing in roots and leaves of wheat varieties ZM9023,JM17,XY54 and YN202 under the condition of phosphorus deficiency,and the gene expression was callback after fill phosphorus,which showed that TaCP genes may play a role in the mechanism of phosphorus stress by regulation of phosphorus nutrition conditions in these wheat varieties.The TaEF gene expression and physiological function under phosphorus stress system was analyzed,and TaCP gene expressed in different wheat varieties of phosphorus stress situation were preliminary analyzed,it may lay the foundation to clarify the role of the gene in plant phosphorus control network.
