Nude mice with hair keratinization anomaly and T lymphocyte immunodeficiency caused by abnormal thymus development have become an important animal model in the field of biomedicine.As female nude mice are unable to bear offspring,Animal Breeding Center often makes nude mice mate with the heterozygote mice,which limits the production of nude mice.Their reproductive performance is apparently diminished by the longer birth interval.Nude mice with more poor digestive function than normal mice,common diet could not meet the needs of their growth development.However,lots of researches already have been done at home and abroad on promoting the growth and reproductive performance of livestock with nutrition control.Yet we still lack special diet fit for nude mice.This experiment aimed promoting nude mice's growth performance through vary levels of protein diet and facilitating their reproductive performance by blend diet with arginine.It also investigated the effect of arginine on the reproductive hormone.1.Effect of different protein level diets on nude mice's growth performanceIn this study,3-week-old Balb/c mice:male nude mice(45),female nude mice(45)and female heterozygote mice(45)were randomly divided into 3 groups(each with 3 repeat,each repeat 5 mice):control group(19%CP),middle protein group(24%CP),high protein group(29%CP),experimental period 5 weeks.At the same time every week,weighed the body weight of 3,4,5,6,7,8 weeks old mice.Test results showed that,the body weight and ADG of male/female nude mice and female heterozygote mice in middle protein group was significantly higher than control group(P<0.05);the body weight and ADG of female nude mice and female heterozygote mice in high protein group was significantly higher than control group(P<0.05);but the body weight of high protein groups were lower than the middle groups without significantly.This experiment showed that 24%CP diet as a well-balance nutrition can advances the growth performance of nude mice.But the lower or higher level of CP will suppress the growth performance of nude mice.2.Effect of diet with arginine on the performance and hormone of reproduction of nude mice3-week-old Balb/c mice:male nude mice(32)and female heterozygote mice(32)were randomly divided into 2 treatment groups:control group(24%CP)and arginine group(24%CP+1%Arg).At 8 weeks old,the same treatment group mice were treated by the combination of male and female 1:1.The statistics contains,mated within 13 weeks,total litters,average birth interval,average litter size,homozygous rate and average weaning weight index.Blood,testicle,epididymis samples were collected at 15-and 21-week-old.The results showed that compared with the control group,the arginine significantly shortened the nude mice of the first and second birth interval(P<0.05).The total litters increased by 34.2%.Also compared with the control group,the average weaning body weight of male/female nude mice and female heterozygote mice was significantly increased by 18.8%,20.5%and 5.7%(P<0.05).The sperm density and testis index of the 15-week-old nude mice in arginine group were significantly higher than control group(P<0.05).But the sperm density and testis index of the 21-week-old nude mice in arginine group were no significantly difference than control group.Compared with the control group,the level of reproductive hormone of the 15 and 21 weeks old male nude mice in arginine group had a trend of increase without significantly difference.This experiment showed that arginine can increases the productive performance of female mice by shortening the average birth interval and accelerates the development of testicle of male nude mice during the growing period.3.Effect of diet with arginine on blood routine parameters and serum biochemical parameters in nude mice3-week-old Balb/c mice:male nude mice(32)and female heterozygote mice(32)were randomly divided into 2 treatment groups:control group(24%CP)and arginine group(24%CP+1%Arg).Blood sample and liver were collected at 21-week-old.The blood routine parameters results showed that compared with the control group,the WBC and ly#of female heterozygote mice in arginine group increased significantly(P<0.05).The serum biochemical parameters results showed that compared with the control group,arginine significantly lowed the level of ALT in male nude mice and the level of ALT and AST in female heterozygote mice and increased the level of UA in the male nude mice and the level of GLU in female heterozygote mice(P<0.05).The antioxidant indexes showed that arginine group significantly raised GSH-Px level and lowed the level of MDA in liver tissue(P<0.05).This experiment showed that arginine could increase the antioxidant capacity of nude mice.
