Spodoptera litura F.is an important pest of crops such as grain,cotton,tobacco,vegetables and so on.The main factors are the high fecundity,predation,omnivorous,overlapping and migratory.S.litura with long-range ability to move,which is the main reason for the outbreak of the disaster,but also the current control of difficulties ofS.lituraconstraints.The aim of this study was to explore the molecular mechanism of flight of S.lituraby the study of flight protein gene of S.litura,and to provide ideas for the prevention and treatment of S.litura.In this study,the full length mRNA sequence of S.liturawas obtained by PCR and RACE.The sequence was 805bp in length and 483bp in open reading frame(ORF),which could encode a protein with 160 amino acid residues.The flightin of S.liturawas highly homologous(>75%)with the flightin of Lepidoptera,but the homology of insects such as Hemiptera,Diptera was low(<40%).The relative expression of flightin in the developmental stages of S.liturawas detected by real-time fluorescence quantitative technique.The results showed that flightin was expressed in larvae from larvae to adult stage,and the expression of flightin was low in pupae stage.The expression level of adult feathers reached the highest value.The relative expression of female females was higher than that of males.The relative expression of flightin in different tissues after Od,Id and 2d of male and female adult worms was further detected.The results showed that the relative expression of flightin in three days after emergence was:head>chest>reproductive system.In the females or males,the expression of flightin was highest at 0 d,followed by rapid decline.In the female reproductive system,the expression of flightin was the lowest at 0d,but increased significantly in the next two days.In the male adult reproductive system,the expression of flightin was maintained at the same level three days after emergence.Furthermore,the efficiency of adult flight protein interference was studied by dsRNA feeding and injection.The results showed that all adults were not eclosion after feeding the dsRNA.Injection of 5?g and 7.5?g flightin dsRNA successfully disrupted the expression of flightin.The expression of flightin decreased after 5?g treatment,31%in 5?g treatment,39%in 5?g treatment,50%in 7.5?g treatment,and 51%in 7.5?g treatment.In this study,the full-length mRNA sequence of S.litura was obtained for the first time.The expression of flightin was found to be low(1d,2d)in the adult chest,and the expression of flightin was consistent with the migration behavior.(1d,2d)model(which was opposite to the change in the expression of chest mass)in the reproductive system of the female,and the expression level of the reproductive system in the female reproductive system was the highest(0d)And the sexual maturity and reproductive behavior of the moth was positively correlated.This study is innovative,results provide new insights into gene function studies of S.litura flightin.
