In this paper,based on the laboratory of Haliotis diversicolor transcriptome,network NCBI and KEGG database,the candidate genes related to energy metabolism of abalone larva were filtrated.The regulation network of energy metabolism was constructed.After RNA interference experiments and induction experiments on larvae of H.diversicolor,These genes were detected by qPCR and ISH.The main research results are as follows:(1)Basing on thyroid hormone signaling pathway and insulin/insulin-like growth factor signaling pathway genes,combining with the NCBI nucleotide databases and the information online,186 candidate genes with clear functional annotation were gotten.These genes were divided into cellular component,biological process and molecular function three categories.Further analysis of the expression level of each gene trends,106 genes s were classified as immune or stress response,protein translation,cell metabolism,cell cycle,membrane proteins,transcription regulation,chromosomal composed of calcium binding,nervous system and unknown nine categories.(2)The correlation coefficient between genes was calculated by SPSS statistical software.The genes expressions and their correlation coefficients were imported into Cytoscape software.The regulatory network of energy metabolism was constructed.This is an attempt for using explosive increasing sequence data.(3)The results of RNA interference experiments and induction of experiments showed that thyroid hormone can positively regulate the MAPK(ERK),PI3 K,HIF-1?,STAT1,metabolism of sugar solution to the original production,sugar fermentation.Moreover,the expression of GSK did not change in the interference experiment,only changed in the induction of the experiment and the opposite trend with thyroid hormone.It suggests that there may be a negative relationship between the pathway and the regulation of thyroid hormone.The sptatial and temporal expressions of TR?,ATP6 showed TR? is mainly located in the dorsal of veligers visceral mass,near muscle pulls base;ATP6 also has a high concentration in TR? high level places.This suggests high expression of TR? may promote the synthesis of ATP,and TR? is involed in the regulation of matter energy distribution.(4)The effects of insulin on energy metabolism not only promote energy metabolism,but also limit the speed of the energy metabolism.Insulin has less extent on energy metabolism than that of thyroid hormones.The results of MIRP,ATP6 ISH showed that MIRP gene is alsolocated in the dorsal veliger visceral mass and near muscle pulls base position,which is very similar to TR?.In addition,there is high expression in foot and velum,after insulin inducing.High or lowinsulin concentration may disorder the energy metabolism of larvae,and lead to high mortality.
