Compound Zhongjiefeng Particles is a veterinary preparation developed both by Guangxi University and Guangxi Dipper Animal Health Products Co.,Ltd.In our study,different extraction methods of Zhongjiefeng were optimized and compared to provide reference to improve the production process.Besides,to offer scientific basis for the clinical application of Compound Zhongjiefeng Particles,a series of experiments were also conducted to explore the effect of Compound Zhongjiefeng Particles on eliminating phlegm and relieving asthma and safety application on target animal chickenMethods:(1)Microwave extraction and ultrasonic extraction of Zhongjiefeng were optimized by single factor experiment and orthogonal experimental design to find out the best extraction of Zhongjiefeng,compared with traditional extraction.(2)The effect of Compound Zhongjiefeng Particles on eliminating phlegm and relieving asthma were carried out in mice and SD rates.(3)Safety evaluation of the Zhongjiefeng were performed in target animal chicken.The one time,three time,five times of the recommended dose of the compound Zhongjiefeng particles were applied on the target animal chicken for 7 days to observe the the effect on clinical appearance,growth performance and blood biochemistry indice.Results:(1)The optimum microwave extracting process conditions acquired by orthogonal experimental design were as follows:15 min of extracting time,80%of alcohol,1:20 of solid-to-liquid and temperature at 70 ?,while the average content of isofraxidin and rosmarinic acid were 0.578 mg/g,0.343 mg/g respectively.The optimum ultrasound extracting process conditions acquired by orthogonal experimental design were as follows:60 min of extracting time,70%of alcohol,1:15 of solid-to-liquid and ultrasound power at 450 W,while the average content of isofraxidin and rosmarinic acid were 0.535 mg/g,0.305 mg/g respectively.The average content of isofraxidin and rosmarinic acid of Zhongjiefeng reflux extraction proceeds were 0.385 mg/g,0.195 mg/g.(2)The mouse model of respiratory excretion of phenol red and rats asthmatic model were successfully established.Compound Zhongjiefeng Particles had a good effect on promoting the amount of phenol red secreted from murine trachea and reducing the level of white blood cell counts and eosinophils.Compound Zhongjiefeng Particles inhibited production of IgE,IL-4,IL-17 and increased the IFN-? level.(3)According to the findings of the target animal safety evaluation,when compared with blank control group,the one time,three time,five times of the recommended dose of the Compound Zhongjiefeng Particles showed no significant difference(P>0.05)on chicken clinical appearance,growth performance and blood biochemistry indice.In conclusion:(1)Among three extraction methods of Zhongjiefeng,microwave extraction showed more advantages than ultrasonic extraction and reflux extraction.(2)Compound Zhongjiefeng Particles had a good effect on eliminating phlegm and treating asthma.(3)Compound Zhongjiefeng Particles showed no any side effect on chicken and was safe in clinical application.
