Abiotic stresses,such as drought,salt,low temperature and biotic stress,such as pathogens and insects,threaten the survival of plants and the production of food,which directly threatens the survival of human beings.In this paper,a new super cold winter rapeseed cultivars Brassica Longyou 6 and weaker cold winter rapeseed cultivars Brassica Tianyou 2 of two Brassica campestris L.as experimental materials,by real-time fluorescence quantitative PCR(RT-RCR)technology research on abiotic stress in Brassica campestris L.Rboh gene family in RbohC/F gene expression and effect of reactive oxygen species the accumulation and activity of antioxidant enzymes,and analyze the relationship between regulation of Brassica campestris L.RbohC/F gene,active oxygen and antioxidant enzyme activity with the MAP kinase cascade pathway,and plant resistance to provide a theoretical basis for the research.The experimental results are as follows:1,Brassica campestris L.Longyou 6 and Tianyou 2 in different tissues by real-time fluorescent quantitative PCR(RT-PCR)showed that the expression of RbohC/F gene in rapeseed Longyou 6 and Tianyou 2 of root and stem and leaf and hypocotyls,the expression of RbohC/F gene in different tissues for the constitutive expression and differences.After low temperature(4?),salt,drought,H2O2 and ABA stress,the expression of RbohC/F was induced in the two kinds of rapes.Prolong with cold stress time,the increase of salt concentration and the increase of exogenous H2O2 concentration,the expression of RbohC/F gene in Brassica campestris L.showed a trend of increased first and then decreased;With increase concentration of PEG-6000,the expression of RbohC/F gene in Brassica campestris L.showed a trend of increased;With increase concentration of exogenous ABA,the expression of RbohC gene increased in Longyou 6;In Tianyou 2,the trend of first rised and then decreased;the expression of RbohF gene increased first and then decreased in Longyou 6,increased in Tianyou 2.By MAPKK inhibitor U0126,H2O2 scavenger DMTU,NADPH oxidase inhibitor DPI and IMD pretreatment after drought and salt treatment showed that the expression of RbohC/F gene in rapeseed were lower than the expression of drought and salt stress,the expression of MAP kinase cascade pathway may be activated by H2O2 and involved in the activation of RbohC/F gene in Brassica campestris L.2,With the increase of salt concentration,two kinds of rapes in the accumulation of H2O2 was the first rised and then decreased trend,the accumulation of O2-was the first decreased and then rised trend;the activity of antioxidant enzymes(CAT,SOD,APX,GR)were the first rised and then decreased trend.With different concentrations of PEG-6000 stress,two kinds of rapes in the accumulation of H2O2 increased and O2-accumulation the downward trend;CAT,SOD,GR activity increased,APX activity was first rised and then decreased trend with increasing the concentrations of PEG-6000.By MAPKK inhibitor U0126,H2O2 scavenger DMTU,NADPH oxidase inhibitor DPI and IMD pretreated by drought and salt stress,antioxidant enzymes(SOD,CAT,APX,GR)activity was lower than that of the salt and drought stress;treated with U0126 before salt and drought stress,the accumulation of H2O2 was higher than that of the salt and drought stress.The result shows that the MAP kinase cascade pathway regulates ROS content by regulating antioxidant enzyme activity,regulation of RbohC/F gene expression by ROS activation of the MAP kinase cascade pathway,further increase the content of ROS.
