In recent years,the popular trend of swine fever in China changed significantly,a wide range of small scale,"atypical" problems,persistent infection and latent infection,especially pig sustained virus infection is the most headache problem.The effective way is vaccination,therefore,vaccine quality directly decides vaccination success or failure of the antigen preparation is the key factor to determine the quality of the vaccine.The purpose of this experiment was to use shaking bioreactor for preparation of CSFV ST cell vaccine,the antigen titer has been further improved.The research content includes the following three aspects:1.The experimental study on the proliferation of ST cells and CSFV was carried out,and the toxin production model was established for the next stage bioreactor.When the CSFV was proliferating in the flask,the titer of 2?3 virus titer reached the peak.2.ST cells and swine toxigenic culture experiment on the application of shaking bioreactor.In the application of shaking bioreactor culture of CSFV experiment,by improving the number of cells per unit volume can effectively enhance the antigen titer.Two kinds of process from the unit volume on the cell number comparison,turn flask culture cell number can reach 2.5 x 105/mL,the reactor is 1.5 x 106/mL,the cells cultured reactor number is 6 times turn the bottle;Compared from the aspects of virus drops degree,reactor than flask culture antigen titer rise by 50%.Through technology improvement,late antigen titer there is room for improvement.
