Tripterygiumwilfordii is a valuable medicinal plant and has high economic value.However,most of the medicinal plants in the planting process,T.wilfordii also suffered from the autotoxicity problem.There are various types of endophytic fungi in the medicinal plants,and endophytic fungi has a great influences on host growth,resistance and secondary metabolism.In this study,anendophytic fungi NS-5 was isolated from Tripterygium Wilfordiiand then constructed the fungal and host symbiotic system.On this basis,with the method of tissue culture,the soil cultivation and cell suspension culture,combining culture with fungal host cell suspension with fungal elicitor addition technique,studying on the endophytic fungi on host generated from the influence of the type and release of autotoxic substances,methods,analyzing in the autotoxicity response of the host and the role of regulation effects,the main results following:Tripterygium wilfordii Hook,f,culture seedling with the endophytic fungi NS-5 that separates from healthy Tripterygium wilfordii plants previously structure a symbiont.and the growth condition of Tripterygium wilfordii,chlorophyll content,element absorption characteristics in evaluation of eight organs aqueous extract could inhibit the increase in different degrees of Tripterygium wilfordii weight,both in Tripterygium wilfordii stem elongation and the increasing of the blade.Inhibition effect:Sterile root exudates>sterile leaf aqueous extract>sterile root aqueous extract>sterile stem aqueous extract;Symbiotic root exudates>symbiotic leaf aqueous extract>symbiotic root aqueous extract>symbiotic stem aqueous extract;Symbiotic root exudates>sterile root exudates;Symbiotic leaf aqueous extract>sterile leaf aqueous extract;Symbiotic root aqueous extract>sterile root aqueous extract;Symbiotic stem aqueous extract>sterile stem aqueous extract.The research of endophytic fungi NS-5 effects receptor tripterygium wilfordii seeds on the autotoxicity.After comparing various indicators like eight tripterygium wilfordii seeds handled germination rate,germination rate,soluble protein,soluble sugar,the enzymeactivity indicators and so on.Compared to the control,the germination rate,germination speed and activity through tissues aqueous extract and root exudates liquid sample handling has decreased,while the various kinds of organic matter of the seeds is far higher than that of the control,which would explain a strong self-poison effect on seed germination tripterygium with various treatments.Analyzed with GC/MS method of liquid ingredients,it is concluded that NS-5 main content,by influencing the autotoxic substances matters to have effect on the autotoxicity of tripterygium wilfordii.Endophytic fungi of tripterygium wilfordii NS-5-induced can strengthen autotoxicity on receptor tripterygium plantlets,might be the tripterygium endophyyic NS-5 elicitor can promote tripterygium wilfordii since the suspension cells release autotoxic substances,and microbial-inducer tripterygium wilfordii suspension cells release ability is greater than the bacteria from the toxic matters inducer tripterygium wilfordii suspension cells released.When time goes by,triptolide suspension cells released autotoxins will accumulate in the culture,it makes autotoxins 9d sample solution containing a higher concentration,so 9d treated plantlets growth of tripterygium<6d treated plantlets growth of tripterygium<3d processing tripterygium plantlets growthof tripterygium.That is:comparing the same time in different sub-inhibitory effect induced with CK-3d<JT-3d<JY-3d;CK-6d<JT-6d<JY-6d;CK-9d<JT-9d<JY-9d;comparing the inhibitory effect of different elicitors at the same time,there is CK-3d<CK-6d<CK-9d;JT-3d<JT-6d<JT-9d;JY-3d<JY-6d<J Y-9d.
