Research On Aneuploidy Of Basidiospore Of Volvariella Volvacea

Using SV primer and tip hyphae isolation and purification,we explored the aneuploidy of spore of V.volvacea.The major results are listed as follows:According to the sequencing data of PYd 15 and PYd21,Zhang Xiaoyin developed 102 SV molecule markers,and constructed a genetic linkage map with 10 linkage groups.According to genome sequence alignment between PYd21 and V23-1,A factor was located in Group 2 by Mauve.H1521 was mated by PYd15 and PYd21.After H1521 was fruited,we isolated 105 single spores.Using SV primer and tip hyphae isolation and purification,aneuploidy of 105 single spore strains were detected.There were 77 haploid single spore strains,other 28 strains could be amplificated two bands by 25 SV markes,in the 28 strains,7 aneuploidy single spore strains V047、V018、V019、V023、V058、V077、V065,6 strains with double A factor.V018×76-7,V018×77-1 was respectively V018 mating with 76-7,77-1.This study verified the presence of aneuploid in single spore of V.volvacea,we propose two assumptions about the appearance of aneuploid.Firstly,because of asymmetric division in meiosis,the nucleuswhose number of chromosome are n+1,n+2 are formation,and they enter basidiospores.Secondly,because of asymmetric division in mitosis,the nucleuswhose number of chromosome are n+1,n+2 are formation,and they enter basidium,then there chromosome were transfered to basidiospores by meiosis.There are no reports about aneuploidy in edible fungi.Study about aneuploidy can enrich geneticKnowledge about V.volvacea,improve the understanding of genetic background.Study about aneuploidy will also play an important role in gene mapping,comparative genomics research and breed improvemen.
