Magnaporthe oryzae is the causal agent of rice blast disease throughout the world,it is a model fungal pathogen for understanding the mechanism of plant-fungus interaction.M.oryzae secretes various of extracellular enzymes,which are likely to be mainly involved in the infection process.A lipase gene FGL1 in Fusarium graminearum has been confirmed to be related to the pathogenicity,while the biological function of lipase in M.oryzae is still largely unknown.Thus,to study a lipase gene MoLl in M.oryzae,we constructed its knockout mutant by homologous recombination method.Our results showed that MoLl deletion mutants were deficient in mycelia growth and conidiation,but their pathogenicities are normal.Subcellular localization analysis exhibited that MoL1-GFP was localized in the appressoria and invasive hyphae during the infection process.Taken together,Lipase MoLl plays an important role in vegetative growth and conidiation process in M.oryzae,while whether it is involved in pathogenesis still needs further verification.In addition,we purified His-tagged MoLl from the culture supernatant of Pichia pastoris through Ni-NTA affinity chromatography,which not only proved that MoLl is a secreted protein,but also established the foundation for further identification of its biochemistry activity.
