Yellow rust is one of the most detrimental diseases of wheat.lt has been proven that breeding resistance cultivars is and will be the most economic,secure and environmental friendly way to defeat the disease spread.The deep investigation of resistance genes in wheat provides solid foundation for disease resistance cultivating.In this study,we cloned TaSYP71 and TaVAMP714 which might be related vesicle trafficking and studied by bioinformatics analysis and functional analysis.Main conclusions are as follows.Sequence analysis indicated it encoded 269amino acid residues containing SYNTAXIN,a conserved domain and a transmembrane domain.Real-time RT-PCR assays displayed that TaSYP71 exhibited up-regulation expression tendency in both compatible and incompatible interaction of wheat and stripe rust fungus in which levels were similar.Subcellular localization assays showed that TaSYP71 was localized in the epidermal cell membrane of tobacco.Yeast overexpression demonstrated culturing cells that contained TaSYP71 expression plasmids enhanced resistance to H2O2 stress in thiamine-deficient medium.What was demonstrated in transient tobacco overexpression assayswas that TaSYP71 inhibited PCD triggered by a mouse Bax protein.Knocking down expression of TaSYP71 in virus induced gene silencing(VIGS)assays impaired the resistance of wheat cv.Suwon 11 to rust disease.These results suggest that TaSYP71 plays roles in wheat-stripe rust fungus interaction and enhances resistance to H2O2 stress,performs as a negative regulator of PCD as well.By combining homologous cloningand in-silicon cloning we obtained the full-length cDNA of TaVAMP714.The gene contained a 669bp open reading frame(ORF)in length,encoding 222 amino and a Longin motif and a Synaptobrevin motifChallenged by exogenous phytohormones treatments,TaVAMP714 wasup-regualted much more by ABA than by ETH and MeJA.While it was up-regulated by PEG and NaCl Wound and Cold treatments could not induce increased expression yet.TaVAMP714 was not strongly up-regulated by different kinds of abiotic stresses.When challenged by stripe rust pathogen,TaVAMP714 was obviously up-regulated in compatible interaction,indicating that they might play important roles in wheat's resistance to the rust CYR31.
