| Endophtic fungus can alleviate various adversity environment and promote the resort to plant growth and development,its role has been confirmed by our predecessors.Aleurites Montana is an ideal tree specie for the biological oil.To promote the growth and development of soil in the south,it’s very necessary to study in endophytic fungi of Aleurites Montana.In the present work we separate and purify the endophtic fungus in different organ parts of Aleurites Montana by tissue separation technology.Select dominant fungus and phosphate-solubilizing fungus and use 18SrDNA to identify these strains.Return inoculation treatment of endophtic fungus conduct in common and low-phosphorous conditions respectively and endophytic fungus form the symbionts with Aleurites Montana.By measuring seedlings height and ground diameter,chlorophyll relative content of leaves with Hand-held portable chlorophyll meter(SPAD-502),chlorophyll fluorescence parameters using fluorescence imaging Handy Fluor Cam,plant nutrients and the content of malondialdehyde(MAD)to analyse the promotion effect of endophytic fungus on host plant growth.1.The plants sample,root,stem and leaf of 29 Aleurites Montana trees in 11 provenances are sampled from Huqiao scattered trees(Bamboo,fir and Aleurites Montana mixed forests)forest,Waidun forestry cutting and cultivating farm and Shufang tree farm in Jianyang,Fujian province.155 strains are acquired after separating and purifying operation.The largest number of strains are acquired from stem organ,60 strains,followed by leaf,50 strains and then the roof,45 strains.2.Endophytic fungus role in the growth-promoting effect of Aleurites Montana(1)The effect of Endophytic fungus on Aleurites Montana biomassThe data of ground diameter,seedling height,root cap ratio,biomass and other indicators show that Pestalotia strains ZJ2 have the most significant growth-promoting effect on seedlings among the selected endophytic fungus.(2)The effect of Endophytic fungus on the photosynthesis of Aleurites MontanaThe data of chlorophyll relative content,chlorophyll fluorescence parameters and other indicators show that Trichoderma strains ZY3 strengthen the photosynthesis ability of seedlings to the maximum extent among the selected strains.(3)The effect of Endophytic fungus on nutrient elements of Aleurites MontanaThe result of C\N\P elements analysis shows that Coniothyrium strains ZY5 are most conducive to the increase of N\P\K content in pants.3.Endophytic fungus role in Aleurites Montana growth under low phosphorus stress.The effect of Endophytic fungus on Aleurites Montana biomass under low phosphorus stress:From the ground diameter and seedings height measurement and analysis we find that the Penicillium strains SJ4 have the most significant growth-promoting effect on seedlings height and ground diameter among the four strains of phosphate-solubilizing fungus in various low phosphorus stress gradient.The data of root-shoot ratio indicators show that both Penicillium strains ZG3 and Thermomyces strains SY1 can obviously promote the growth of mot system in various low phosphorus stress gradient.The analysis of plant biomass in various low phosphorus stress gradient indicate that the Penicillium strains SJ3 show strong ability to relieve the impacts of stress.Comparing all quotas,the strains SJ4 have better capabilities to relieve the stress of low phosphorus and accumulate the plant biomass than others.4.Endophytic fungus role in Aleurites Montana physiological ecology under low phosphorus stress.The effect of Endophytic fungus on the photosynthesis of Aleurites Montana under low phosphorus stress:The data of chlorophyll relative content,chlorophyll fluorescence parameters and other indicators show that all of the four strains phosphate-solubilizing fungus have capabilities to relieve the stress of low phosphorus and promote the plant growth,strengthening the photosynthesis ability of plants.The SJ4 strains show the most obvious effect on plants.According to the MDA content in plants Penicillium strains including SJ4,SJ3 and ZG3 help decrease the injured degree of cells.5.Endophytic fungus effect on nutrient content in Aleurites Montana under low phosphorus stress.Based on the determination of carbon content,nitrogen content in plant aerial portion and underground part,higher C/N ratio in plant aerial portion is observed generally.The data of phosphorus content in plant aerial portion and underground part show that Penicillium strains ZG3 can significantly promote the increase of phosphorus content,especially under mild,moderate or severe stress.And the data of K content in plant aerial portion and underground part show that Penicillium strains SJ4 can promote the increase of K content.While it’s obvious in plant aerial portion under normal condition and mild or severe stress,it seems that SJ4 promote the increase of K content significantly in underground part under normal condition and mild stress. |
