Tomato(Lycopersion eseulenrum Mill.)is annual or perennial herbaceous plant in the Solanaceae Lycopersicon,the ripe juicy berries are the products.It's rich in nutrition,good taste,and ate by a crow of people,and is also regarded as fruit in vegetables.Tomatoes are rich in lycopene and vitamin C,they play a very important role in antioxidant,prevention of cancer and other aspects.This thesis studied on the physiological,biological and nutritional functions of wild tomato from spectroscopy and nutrition aspects,researched on the construct and expression level of p7 gene from the molecule biology,and compared with cultivated tomato.Not only have theoretical significance,but also provide the basis in the exploitation and utilization of nutrient-rich wild tomato.The results of the experiment are as follows:(1)Comprehensive analysis and comparison of wild tomato nutrients.It was found that vitamin C,lycopene and total acid content of wild tomato was high,amino acid was rational;and wild tomato was rich in calcium,manganese,magnesium,iron,sodium and other mineral and trace elements;and was of higher potential value in nutrient.Compared with the cultivated tomato,vitamin C,lycopene,flavonoids,amino acid,pectin,Ca,Fe,Mg were significant changes.The wild tomato vitamin C content as high as 92.71mg/g,almost 30%higher than the cultivated tomato;lycopene content was also up to 0.25 mg/g,almost 1 times higher than that of the cultivated tomato;and tyrosine,lysine and proline contents was more than 1 times higher than the cultivated tomato.It was show that wild tomato was more high nutritional value than cultivated.(2)The mechanism of antioxidant capacity of wild tomato was discussed.The results showed that the enzyme activities of SOD,CAT,POD and GR were higher than the cultivated tomato,only the activity of APX was slightly lower than cultivated.Compared with cultivated tomato,the wild tomato POD activity was higher above 3 times than cultivated,reached 1.02U·g-1·min-1;GR activity was more thanl times,its activity was up to 4.11 U·g-1·min-1;SOD and CAT activities were more than 20%higher,and they reached 2.36 U·g-2·min-1 and 7.33 U·g-1·min-1,respectively.The results showed that the ability of wild tomato in antioxidant,anti-aging,as well as maintaining nutritional quality of fruit was more.(3)Preliminary using molecule biology knowledge to construct P7gene.In the analysis of expression patterns of tomato,the experiments showed that the P7 gene under the salt treatment condition,the roots,stems and leaves of P7 were up-regulated expression,and the expression in roots increased sharply,while the MM had no such changes.In the gene construction,the nucleotide sequence of P7 was basically consistent with SGM on the site,and there were only 1 nucleotides difference,the amino acid at 698bp changed from A to B,and caused the mutation at 233bp H to R,resulted in an amino acid histidine to arginine.
