Light is one of the most important environmental factors,which plays an important role in growth,development,morphogenesis and metabolism of plant plantlets in vitro.The phytochrome,cryptochrome and UV-B receptors were light receptor,which monitor signal direction,energy,light quality,and regulate growth and development of plants.LEDs(Light-emitting diodes)are solid non-coherent light emitters,which can convert electrical energy into light energy.LEDs has many advantages,such as small size,long life,low energy consumption,securing wave length and low production of heat,which can emit the exact spectrum based on the need of plants,so that it can be widely used in seedling breeding,vegetables cultivation and plant tissue culture.There are some previous studies in oversea about the affect of growth and morphogenesis at plantlets in vitro by LEDs.But little known about the effect of crop seedlings and plantlets in vitro in the domestic.Spectral light changes evoke different morphogenetic and photosynthetic responses that can vary among different plant species.Many studies about growth and development were done in recent,but many questions about effects on high plants still were not understood.And there have no reports about the effect of potato by LEDs.This study was to determine the effects of different spectrum on the growth and minituber induced of potato plantlets in vitro,containing two potato varieties:Shepody(SP)and Favorita(FT),and select the feasibility of application of LEDs light used in the growth and minituber induced of potato plantlets in vitro.Main research results were as follows:1.The effects of different light-emitting diodes(LEDs)lights on growth and morphogenesis of potato plantlets in vitro were invested.The results showed that,stem diameter,specific leaf weight(SLW),dry weight(DW)and health index of potato plantlets in vitro were significantly higher under the combined light of blue,red and yellow(RBY)than those under the combined light of blue,red and green(RBG);the leaf area was larger under RBG than that under RBY;stem diameter,SLW,fresh weight(FW),DW and health index were significantly higher under the combination of spectral contained red light at 660nm than those under the combination of spectral contained red light at 630nm;the DW and health index were significantly higher under the combination of spectral contained blue light at 445nm than those under the combination of spectral contained blue light at 465nm.2.The effects of different light-emitting diodes(LEDs)lights on Physiological and biochemical of potato plantlets in vitro was invested.The results showed that the content of chlorophyll,soluble sugar and starch were high under the combination of spectral contained green;the soluble sugar and starch were significantly higher under the combination of spectral contained red light at 660nm than those under the combination of spectral contained red light at 630nm;the soluble protein and starch were significantly higher under the combination of spectral contained blue light at 445nm than those under the combination of spectral contained blue light at 465nm.3.The effects of different light-emitting diodes(LEDs)lights on stomatal traits and anatomical features of leaf of potato plantlets in vitro was invested.The results showed that the stomata density of potato plantlets in vitro was greatest under R660B445Y,however,the stomata size was large under R660B445Y;stomata density of potato plantlets in vitro were greatest under FL,and the stomata size was small.In the RBY treatment,a clearly defined upper epidermis occurred in the leaves.In addition,the palisade cells in the RBY treatment were neatly arranged.The intercellular air spaces among the spongy mesophyll cells in potato leaves were smallest under RBY.4.The effects of different light-emitting diodes(LEDs)lights on minituber induced of potato plantlets in vitro was invested.The results showed that LEDs were contributed to increase the photosynthetic product and the accumulation of biomass,and have a major impact to the microtubers induced on the latter.The blue light at 445nm can promote tuber rate and tuber quality.The blue light at 445nm and yellow light facilitate the microtubers induced on the latter.In conclude,the combination of red,blue and green LEDs were the best lights for the growth and microtubers induced of potato plantlet in vitro.The red light at 660nm was conducive to growth of potato plantlet in vitro.The blue light at 445nm was not only to strengthen the seedling,but also play an important role for the induction of potato plantlets in vitro.In the whole experiment,the treatment of LEDs is better than fluorescent.Then,LEDs can be widely used in the potato for strengthen plantlets in vitro and microtubers induced.
