Due to long single germplasm resources,Zhangzhou local Auricularia polytricha has gradually degenerated and hybrid is essential to enrich the resources.By single spore cross breeding,this study selected a mutation AP2010,as the parent,crossbreeding with and Zhangzhou local species 4328,43012 and AP201.This study mainly contains the following parts:1.This study identified the selected 4 species by morphology and ITS sequencing which phylogenetic tree was established.Compared cultivation characters of 4 species after fruiting experiment,these species are obviously different.It could bring rich germplasm resources of hybrid-breeding by hybrid 4 species with each other2.We made an accurate determination of mating factor for the mononuclear strains using conventional mating type determining methods and analysed by chi-square test.The result that the polarity is the bifactor hetenthallism was similar to recent research.And we also presumed that 4 strains had 4 A factors and 4 B factors,which proved that five parental strains had abundant genetic diversity.3.We observed clamp connection and antagonism reaction,combined with the method of RAPD molecular markers and finally screened 23 F1 strains.Compared mycelium growth rate of 23 F1 strains on PDA and on cultivation material,6 and 9 strains were higher than their parents respectively.Regression analysis was carried out on the two groups of data,combining ISSR clustering analysis map,hybrid strains growth rate were related with parental strains.We compared the data of yield and morphological characteristic after fruiting experiment and found 6 F1 strains were higher than parents in yield and AP2010-4/43012-12 had better morphological characteristic.
