Establishment Of Animal Model Of Source Bovine Neospora Caninum Infect The Chicken

Neospora as a kind of newly discovered intracellular parasites.Its parasitic hosts have been extended all the time.At the beginning,it can only led to the infection and death of dogs,then,it founnded that the canine is the final host,including dogs,coyotes and dingoes.Next,at the clinical,bovini were founded that they were the most susceptible animal in ruminants,which opened the prelude of Neospora infected ruminants.From the carnivorous canine,to the grazing ruminants,the field of neospora infection has been expended gradually.Although,there is no evidence suggests that Neospora can infect humans,studies have shown that antibodies of neospora has ben found in human serum.And neospora can also invaded into a variety of anthropogenic cells in vitro culture,so,we can not ignore the possibility of neospora would infect humans.Chicken,as the important source of food and nutrition of human and other animals,Studies have confirmed that neospora have been found in the sparrows and adult chicken and pigeons.In consequence,the hazard area of Neospora endanger ranges widespread.This research select chicks for the experimental animals,used the isolates of neospora to infect chicks to establish the animal model of neospora infected chicks,to make sure the pathogenicity of Neospora infected chicks.For the experimental group,five doses group chicks were given the neospora tachyzoite at 10,102,103,104,105 respectively used intraperitoneal inoculation,normal control group were established at the same time,observed the clinical symptoms,collect stool and record every day;pathological observed the organization'sdamage in brain,liver,spleen and other organs of ovaccination chicks.Observed the insect body invasion pathways in immunohistochemistry.Detected the polypide DNA of each viscera tissue,blood and stool in PCR.The results show that after infected the Neospora,they appeared some clinical symptom,such as hair disheveled,diarrhea,depressed of spirit,arthrocele and incoordination,even accompanied by death;In pathology HE dyeing,we found that each viscera tissue extensive hemorrhage,solid organ degeneration necrosis,of which,liver,lung,intestine and brain lesions were the mostobvious;In immunohistochemistry,we have found the positive area of worm antigen,but did not see clearly the neospora tachyzoite existence;PCR detected the insect body DNA in the brain,liver,kidney,lung and other organs,organizations and faeces.The research establish the animal model of neospora infected chicks successfully,made a foundation for the research of neospora pathogenic role of poultry.
