In recent years,the abuse of antibiotics has caused more and more broad spectrum of drug-resistant bacteria.The most of antibiotic effect is generally reduced.The antibacterial mechanism of traditional antibiotics is mainly to kill bacteria or inhibit the proliferation of bacteria.Under the guidance of the theory of new antibacterial drug development may also be difficult to avoid the production of bacterial drug resistance.Recent studies have found that the virulence of the bacteria is closely related to the virulence of the toxin secreted by the bacteria.It is possible to find a new way to find a new type of antimicrobial agents with the virulence of pathogenic bacteria as the target.Traditional Chinese medicine has a significant effect on the treatment of bacterial diseases.Our previous study found that Niujiao Dihuang decoction has significant inhibited hemolysin induced by hemolysis of red blood cells,Chishao anti hemolysis is the strongest.The anti-hemolysin active components in Radix Paeoniae Rubra were analysised,and the pharmacokinetics of the ingredients in rats was investigated To provide basic data for the research of traditional Chinese medicine components which can inhibit the bacterial virulence factors.The HPLC analysis method of Chishao Aqueous extract was established.And the chromatographic working conditions were chromatographic column C18 column,the mobile phase was water acetonitrile,water as the mobile phase A,acetonitrile as mobile phase B;gradient elution process is as follows:A 95%,10 min;A 95-90%,10 min;90-80%A,10 min;80-100%A,5 min;velocity 1 ml/mim,column temperature of 25 ?,sample size:50 ?l,chromatographic analysis time for 60 min.The method can effective separated Chishao in each component.This method is stable,accurate,high sensitivity,The method is suitable for analysis the composition of Chishao.The chromatographic preparation method was established based on the analysis method above.The hemolytic activity were detected on the product of Chishao.the results showed that the anti-hemolysin composition of Chishao and paeoniflorin have similar peak time.But paeoniflorin has no inhibitory effect of the hemolytic activity of hemolysin.And the heating effect of the extraction process of Chishao can increase its anti hemolysin activity.Chishao extract were analysised by the methods of Infrared spectroscopy,nuclear magnetic resonance(NMR)and mass spectrometry analysis.The results suggest that the anti-hemolysin activity components of Chishao may be a kind of paeoniflorin as aglycone and glucoside linked oligosaccharide glycosyl class of compounds.Drug content detection method in plasma of Rat was established based on the analysis method above.This method was accurate,stable,without interference of endogenous substances,and the recovery rate was 101?107%,the coefficient of variation was 3.95-13.68%.Detection of limit was 35 ng/ml.The pharmacokinetic analysis of the components in rats in vivo Tmax was 0.17 h,Cmax was 11.75±1.35?g/kg,half-life was 0.9289 h,...
