Avian Mycoplasma,including Mycoplasma Gallisepticum(MG)and Mycoplasma synoviae(MS),is one of the most important pathogenic disease in poultry production.The major symptoms for MG infection include rales and coughing,and MS infection is mainly for the joint swelling and decreased hatchability.Avian mycoplasmosis induce low mortality rate in chicken,but it may cause huge economic losses in poultry industry.In order to reduce the influence of Mycoplasma on poultry industry,the Mycoplasma immune baseline of chicken was established in our study to improve the chicken immune monitoring system in farm.In order to ensure the accuracy of ELISA,the operation steps were optimized in this paper firstly.In the course of ELISA,the low temperature preservation,centrifugal treatment and freezing-thawing procedure likely to affect the test results.Our test showed that preservation at-20 ? for one week have no effect on the result of ELISA test,but preservation for four weeks affect the result significantly.The serum antibody levels was significantly reduced after freeze-thaw treatment,and also declined with the increase of the number of freeze-thaw cycle.Centrifugation have no effect on the detection of serum antibody levels.In our study,the serum antibody titer of white feather broiler in three breeding chicken were tracked and monitored by the optimized ELISA for 160 days to establish the MG and MS immune baseline of chicken.The serum samples were collected from chickens of 1 day,45 days,60 days,80days,120 days,145 days and 160 days old.The antibody title was detected with ELISA.The results were analyzed statistically to establish the MG and MS immune baseline of white feather broiler breeder for determining the range as signs of infection,and thus improving the broiler breeder farm immune monitoring system.
