Adjuvants may be added to vaccine to modify the immune response by boosting it such as to give a higher amount of antibodies and a longer-lasting protection,thus minimizing the amount of injected foreign material.Adjuvants may also be used to enhance the efficacy of vaccine by helping to subvert the immune response to particular cells type of immune system,for example by activating the T cells instead of antibody-secreting B cells depending on the type of the vaccine.Adjuvants can be classified according to their component sources,physiochemical properties or mechanisms of action.Two classes of adjuvants commonly found in modern vaccines include immunostimulants and vehicles,whereas some components can combine both properties.Nano adjuvants as a kind of novel adjuvants,can enhance the vaccine effectively with good security,but at present the study of its mechanism of action is not enough.Porcine transmissible gastroenteritis coronavirus(TGEV)is an animal coronavirus that causes severe gastroenteritis in young TGEV-seronegative pigs.Various breeds of pigs,regardless of age,are susceptible to TGEV;however,the mortality rate for piglets under 2 weeks of age is the highest,reaching almost 100%.Diseased pigs often present with vomiting,dehydration,and severe diarrhea.Further,the disease is known to affect pigs in many countries throughout the world and an outbreak can cause enormous losses in the pig industry.For the protection of neonatal and older animals from disease,vaccination is an effective prevention measure.Although,there are many commercial vaccines,the traditional inactivated vaccines have many deficiencies.Therefore,we prepared the two kinds of adjuvants TGEV inactivated vaccines,and study its immune effect on mice.And made a preliminary study of the mechanism of nanometer silicon adjuvant enhanced vaccine immune effect.The concrete research content is as follows:1 We prepared the nano silicon adjuvant TGEV inactivated vaccine and aluminium hydroxide adjuvant TGEV inactivated vaccine,and evaluated its safety.Then evaluate the two vaccine immune mice of its effectiveness.The results show that the two kinds of vaccine is safe for mice,can induce immune response in mice,nanometer silicon adjuvant inactivated TGEV vaccine can induce stronger cellular immunity and humoral immunity in mice.2 24 h and 48 h after immuned with two kinds of TGEV inactivated vaccine,total RNA was extraction from muscles for detection of gene expression profiles.The results showed that nano silicon adjuvant TGEV inactivated vaccines can increases immune related genes such as CD7,CD86,TLR1,TLR4,TLR6,CD40,TLR2,IRF5,IRF7,TLR3,TLR7,OASL1,OAS3NOD2,IRAK4,and immune related go term such as inherent immune and inflammatory reaction,chemokine activation and Pathway such as toll-like receptors signaling pathways.Analysis found that nano silicon adjuvant TGEV inactivated vaccine could significantly induce the activation of innate immune system in mice.3 24 h and 48 h after immuned with two kinds of TGEV inactivated vaccine,total RNA was extraction from muscles for detection of the expression of key factor in immune related pathways with fluorescent quantitative polymerase chain reaction(PCR).The results showed that nano silicon adjuvants can cause IRAK1,IRF5,Myd88,TICAM2,TRAF6,TICAM 1 a significant rise in toll-like receptor pathway in,and can activate the nf-kappa B pathway.Prove that nanometer silicon adjuvant activated the natural immune system and adaptive immunity through toll-like receptors signaling pathways.We prepared the nano silicon adjuvant inactivated TGEV vaccine and aluminium hydroxide adjuvant inactivated TGEV vaccine and verified its immune effect,also has a preliminary study on the mechanism of its enhancement on vaccine immune effect.Results show that nanometer silicon adjuvant inactivated TGEV by activating toll-like receptors signaling pathways regulate various cytokines and inflammatory cytokines produced and released,and then quickly activate the natural immune system and adaptive immunity,to achieve the enhancement purpose to the vaccine immune effect.
