Light Field Microscopy With Light-sheet Illumination

We invented a 3D microscope system of high spatio-temporal resolution.The microscope is of multi functions and multi-mode to meet different imaging conditions.The system has the advantages of both light-sheet fluorescence microscope and light field microscope.By combining the light-sheet illumination and light field detection,we has achieved 3D dynamic imaging in thick samples and broaden the application of the light field microscope.The system is built from a commercial Olympus BX-51 upright microscope.The imaging mode can be easy switched from light-sheet microscope to light field microscopy with original microscope function reserved.The compact and module design make the system establish friendly and easy improvable according to different demands.Up to 100 FPS volume imaging rate is achieved with cellular resolution by the system.We imaged the C.elegans and zebrafish heart with our system and reconstructed high resolution stationary as well as moving procedure of C.elegans,the beating heart of zebrafish in three dimensions.The system is based on real biomedical problems and we want to provide powerful and useful device for such researchers.We intend to provide a new method for microscope community.We hope our system can be better improved and has a widely application.
