As a new technology of tumor ablation,nanoknife has achieved remarkable results in the treatment of pancreatic and prostate tumors by virtue of its advantages of minimally invasive,targeted therapy and non-thermal ablation.The principle of nanoknife is based on the hypothesis of cell perforation under high-voltage pulsed electric field.When the high-voltage pulsed electric field produced by nanoknife is applied to cells,it causes cell membrane perforation,which leads to cell disintegration and death after a certain degree of membrane perforation,thus achieving the effect of tumor ablation.At present,the nano-knife used in clinic is imported,and there is no domestic nano-knife put into clinic.Therefore,the research and development of nano-knife in China is in urgent need of breakthrough.Based on this,the key technology of nano-knife high-voltage pulse power supply is developed,and the mechanism of nano-knife is further explored.To explore the synergistic effect of nanoknife electromagnetic characteristics and targeted therapy through animal experimental modeling.The main research contents and results are as follows:1)In the research and development of key technology of nano-knife high voltage pulse power supply,the circuit design of "Electricity of city input?Rectifier filter circuit--DC-DC circuit--Push-pull inversion circuit?Voltage doubling and rectifier circuit--Pulse generating circuit--Pulse output"is realized in the circuit module.In the control module,the MCU is used as the control core to control the driving signal of the circuit,alarm the circuit and control the output.Finally,the prototype realizes 0-600V adjustable output voltage,500Hz-1000Hz adjustable output frequency,5-42 adjustable output pulses in the pulse group and 10us-50us adjustable output pulse width.2)Based on the oscilloscope Horizontal module,this paper designs a pulse output detection method.The test results show that the errors of pulse frequency,pulse width and pulse number are small,and the errors of output pulse voltage value are relatively large in each group.3)In this paper,magnetic nanomaterials are used as carriers to combine the nanoknife principle with the nano-targeting therapy principle to achieve the synergistic effect of physical therapy and chemical therapy.Fe3O4 solution with corresponding magnetic properties was selected to explore electromagnetic targeting through animal experiments.The experimental results show that the aggregation of Fe3O4 reagents to the tumor site can be enhanced by applying a certain intensity of pulsed electric field to the tumor site,and the intensity,frequency and width of pulsed electric field are proportional to the aggregation effect of Fe3O4 to the tumor site.
