In living organisms Sulfur dioxide?SO2?plays a crucial role in maintaining the redox balance and regulating cardiovascular function in the body.However,excessive SO2and its derivatives can cause great harm to the human body,causing adverse reactions and serious diseases.Therefore,it is of great significance to develop the monitoring of sulfur dioxide and its derivatives.Fluorescent probe has attracted much attention from scientists because of the advantages of the excellent specificity,sensitivity and good biocompatibility.In this thesis,a series of reactive fluorescent probes?X-1,X-2 and X-3?were designed and synthesized,and their spectral properties and fluorescence imaging were studied,so as to achieve a highly selective and sensitive detection of SO2derivatives.The work of this thesis contents are as follows:Chapter 1:This chapter introduces the concept composition and mechanism of fluorescent probes,focusing on the research progress of the HSO3in biological mercaptan fluorescent probe.Finally,the project of this thesis were proposed according to the preliminary work.Chapter 2:The reactive fluorescent probe X-1 with naphthalene-benzothiazole as fluorophore,was designed and synthesized for HSO3.The results showed that the probe could quickly identify HSO3?40 s?by nucleophilic addition reaction.The color of the solution changes from orange to colorless with prople,and that means probe X-1 can be used for“naked-eye”recognition of HSO3.At the same time,the probe showed a high sensitivity,with a detection limit of 0.322?M,and realized the detection and cell imaging of HSO3in living cells.Chapter 3:The reactive fluorescent probe X-2 with Julolidine-TCF as a fluorophore was designed and synthesized for HSO3.The research shows that the probe shows excellent selectivity for HSO3,rapid response?700 s?and high sensitivity?detection limit as low as 0.3?M?.And the color of solution changes from blue to colorless after the response,with the naked eye recognition effect.Chapter 4:A dual-function fluorescent probe X-3 based on benzpyron-coumarin was designed and synthesized for HSO3and biothiol.The results showed that the probe X-3had good optical stability and high selectivity in PBS?p H=7.3,10 m M?buffer solution.HSO3and biothiol?GSH,Cys?could be specifically recognized by different excitation wavelengths?469 nm and 580 nm?,with the colors changed from mauves to orange and blue-green respectively.
