Sargassum horneri are large marine brown algae,which have the characteristics of wide distribution,large biomass,easy cultivation,and rich content of polysaccharides.However,due to the complex structure and toughness of brown algae cell walls,the development of Sargassum horneri resources is hindered and it is difficult to achieve products High value-added utilization.In this paper,Sargassum horneri is used as a raw material,on the basis of the isolation and screening of high-yield alginate lyase strains,and Sargassum horneri is used as a substrate.Release and degradation,and then further separate the components of the fermented Sargassum horneri fermentation broth.The anti-fatigue effects of the full component of Sargassum horneri fermentation broth(SHF-1)and the extract of brown algae oligosaccharide component(SHF-2)and polysaccharide component(SHF-3)were investigated by the exhausted swimming experimental model of weight-bearing mice effect.The research results are as follows:(1)Screening and isolation of high-enzyme-producing strains and optimization of culture conditions:Several strains of alginate-degrading bacteria were isolated from the intestinal tracts of decaying Sargassum horneri and the intestines of Abalone.The strain was named HZ-3 through the determination of enzyme activity and the stability screening of continuous generation of enzyme production.After 16S rDNA sequencing identification and phylogenetic tree construction,HZ-3 was identified as Vibrio alginolyticus strain.Through the single-factor experiment and orthogonal experiment on the fermentation medium of HZ-3 strain.The optimum enzyme-producing medium components of this strain were determined as follows:The addition amount of sodium alginate is 0.8%,the addition amount of peptone is 0.7%,the concentration of NaCl is3.0%,and the concentration of K2HPO4·3H2O is 2.2%.The optimum conditions for this strain were determined:the cultivation time was 42 h,the cultivation temperature was30℃,pH 7.0,and the inoculation amount was 2%.The enzymatic activity of alginate lyase obtained under this condition was 81.94 U·mL-1,which was 1.26 times that of the enzyme activity before optimization.(2)Optimization of Sargassum horneri fermentation process conditions:Taking fermentation time,pH,ratio of material to water and inoculation amount as experimental factors,the production amount of brown algae oligosaccharide is the response index.Using response surface methodology(RSM)to optimize the fermentation process conditions of HZ-3 strain fermentation to degrade Sargassum horneri to produce brown algae oligosaccharide.The results showed that the best process parameters were:fermentation time 56 min,pH 7.1,feed-water ratio 1:6,inoculation volume 10%.Under this process condition,the extraction rate of brown algae oligosaccharides in cuprophyta powder was37.0%.Compared with water extraction,it is 1.56 times that of water extraction.(3)Study on the anti-fatigue effect of the fermentation broth extracts with different treatments:The anti-fatigue effects of the full component of Sargassum horneri fermentation broth(SHF-1)and the extract of brown algae oligosaccharide component(SHF-2)and polysaccharide component(SHF-3)were investigated by the exhausted swimming experimental model of weight-bearing mice effect.The results showed that after 30 days of gavage.Compared with the blank control group,the weight of mice in each experimental group was not significantly different(P>0.05).Indicating that the three copper the algal fermentation broth extract had no significant effect on the weight of mice.It showed that the extracts of the three Sargassum horneri fermentation broths had no significant effect on the weight of mice.The exhausted swimming experimental model of weight-bearing mice showed that the extract of Sargassum horneri fermentation broth has good anti-fatigue effect.Among them,the weight-bearing mice in the SHF-1 medium-dose group had the longest exhausted swimming time.Compared with the blank group,it increased by 105.04%.But slightly lower than the positive group(American ginseng extract).All three fermented extract components can increase liver glycogen(LG)reserves of weight-bearing mice.Reducing blood lactic acid(BLA)and blood urea nitrogen(BUN)levels.Reducing the enzyme activity of lactate dehydrogenase(LDH).Comprehensive analysis of various detection indicators,the anti-fatigue activity of all components of Sargassum horneri fermentation broth is the best.The research results have laid a good research foundation for the development of Sargassum horneri resources as functional food or health products.
