In this thesis,three kinds of chiral silica gel spheres were synthesized by polymerization-induced colloid aggregation(PICA)method,which were used as chiral stationary phases for enantioseparation in high performance liqiud chromatography(HPLC).The main researches are as follows:(1)A HPLC chiral column,composed of L-Glu silica gel spheres,was fabricated by conventional high pressure slurry packing technique.Fifteen racemates and eleven positional isomers were resolved on the column.The good chiral separations of Troger’s base,praziquantel,3-benzyloxy-1,2-propanediol,1,2-epoxyhexane,acetoin,2-methyltetrahydrofuran-3-one and glycidyl isopropyl ether were obtained.The relative standard deviations for five replicate separations of racemates were 1.12 %for retention times and 3.24 % for peak areas.(2)The L-Thr silica spheres was synthesized as chiral stationary phase to separate racemates and positional isomers.The experimental results showed that nine chiral compounds including hydrobenzoin,2,2-diphenyioxirane,3,5-Dinitro-N-(2-phenylethyl)benzamide,4-hexanolide,methyl(R)-(-)-3-hydroxybutyrate,limonene,N,N-dimethyl-1-phenylethylamine,3-butyn-2-ol,1,2-Propanediol and eight positional isomers were resolved on the column.(3)We reported a chiral stationary phase composed of L-Cys silica gel spheres in HPLC.The column was able to recognize fifteen racemates and eleven positional isomers.The effects of separation were also discussed by injected volume of samples,retention times and peak areas.The L-Cys chiral silica spheres is a promising candidate for the chiral stationary phases of HPLC.
