Objective:The seeds of the Althaea rosea,a seed of the Malvaaceae plant Althaea rosea?Linn.?Cavan Althaea rosea records found in the Xinjiang Uygur Autonomous Region Uygur Medicines Standard?2010 Edition,Book 1?.At present,the main purpose of Althaea rosea seeds is medicinal materials.The Althaea rosea seeds has higher oil content and contains more unsaturated fatty acids.According to literature surveys,research reports on the Althaea rosea seeds fatty oil concentrate on fatty acid content analysis.However,there are no reports on the extraction method,infrared characterization,physicochemical properties and oxidation stability of Althaea rosea seeds fatty oil.This article made a systematic study on the fatty oil of Althaea rosea seeds,including the extraction method of oil and extraction process,infrared characterization and the separation and identification of five main components,physicochemical properties and oxidation stability.In order to further expand the pharmacological efficacy of wolfberry fruit,and rationally develop and use wolfberry fruit to lay the foundation.Method:According to the effects of different solvents on the extraction rate and properties of the fatty acids from Althaea rosea seeds oil,the best extractant was determined.The physicochemical properties of different extraction methods and seven batches of Althaea rosea seeds fatty oil were determined with reference to the fourth edition of"Chinese Pharmacopoeia".Althaea rosea seeds fatty oil content determination reference GBT5009.6-2003,using single factor method and orthogonal experiment to determine the optimal technology Althaea rosea seeds oil.Infrared spectroscopy regular pattern of 7 batch Althaea rosea seeds were explored by Infrared spectroscopy.Determination of five Fatty acids in Fatty Oil of Althaea rosea seeds by Pre-column Derivation-HPLC.Determination of the content of Pb,As,Hg,in the fatty oil of Abelmoschus hollyhupii by flame atomic absorption spectrometry and atomic fluorescence spectrometry.The effects of temperature,light,air,metal ions,the acid value of antioxidants on Althaea rosea seeds fatty oil and peroxide value influence.The effect of temperature on the content of five fatty acids in the fat oil of Abelmoschus chinensis was studied by HPLC method.Results:The best extraction solvent for petroleum ether,the best extraction method for ultrasonic extraction.The transparency,odor,refractive index,relative density,peroxide value,acid value,saponification value and iodine value of 7batches of Seeds of Althaea rosea seeds fatty oil were:transparent,no smell,1.47211.4728,0.91340.9199,2.3203.7709?mgKOH/g?,5.568.79?mgKOH/g?,198.10212.78?g/100g?,119.78130.34?gI2/100g?.The optimum extraction conditions of ultrasonic extraction were as follows:ultrasonic power was 200 W,extraction temperature was 30?,extraction time was 30 min,the ratio of material to liquid was 1:10 g/mL,the degree of disintegration was 50 mesh and Extraction times was 1.After optimization,the extraction rate of hollyhock seed oil was 12.57%.There are some differences in the number,shape and intensity of infrared absorption peak in fat oil of 7 batches of Abelmoschus hupiensis.By comparing the infrared spectrum of fat oil,it is found that there are 14 peaks in total.The content range of?-linolenic acid,oleic acid,linoleic acid,palmitic acid and stearic acid in seven batches of fat oil of Althaea rosea seeds is as follows:0.41%1.23%,15.98%19.94%,34.15%53.41%,4.22%7.72%.The range of the content of Pb,Hg,As,is as follows:00.035 mg/kg,4.88×10-40.089 mg/kg,9.30×10-40.03mg/kg.Through the discussion on the stability of the fat oil of the Althaea rosea seeds,it can be seen that the storage of fat oil of the seed is beneficial to the storage of the fat oil under the conditions of low temperature,closed without light and low content of metal ions.And the addition of appropriate antioxidants can prolong the shelf life of the fat oil.Conclusion:In this paper,the extraction method of fat oil from Althaea rosea seeds was studied.The results show that the ultrasonic extraction method is more stable.There were differences in the extraction rate of fat oil and the peak type of infrared characteristic peak in 7 batches of hollyhock seeds.The determination of five fatty acids in the fat oil of hollyhock seeds by HPLC was also very different,which may be due to the different growth environment of the seeds.Caused by the difference in content.The acid value and peroxide value of the fat oil were slightly higher than that of the national standard.The effects of temperature,light,air and metal ions on peroxide value and acid value were discussed.The results showed that extraction temperature,light,air and metal ions all affected the acid value and peroxide value of the fat oil.
