Silica-based mesoporous materials not only have large specific surface area and pore volume,orderly pore structure,narrow pore size distribution,continuous and adjustable pore size,but also are non-toxic,harmless,and good biocompatible,which makes them widely used in the fields of polymer adsorption,biological sample separation,drug storage and transportation.The mesoporous silicas modified with organic functional groups can well meet the needs of modern biological sample analysis,as they combine the huge diversity of organic groups with the porous structure proterties of mesoporous silica skeleton.As a result,the fabrication of these materials opens a new path for the separation and enrichment of biological samples.1.Functionalized mesoporous silica materials:Four kinds of mesoporous silica with different pore size and specific surface area were prepared by sol-gel method.GSH-functionalized mesoporous silicon material was obtained by functional modification of glutathione?GSH?to obtain glycopeptides material as hydrophilic interaction chromatography?HILIC?.The material was used to enrich glycopeptides in the protein hydrolysate of immunoglobulin G?IgG?,the mixture of IgG and BSA digestion,and the difference of mechanism was discussed.Four kinds of mesoporous materials can"capture"2426 glycopeptides from IgG protease hydrolysate with good specificity and sensitivity.In addition,543 independent N-glycosylated peptides derived from 162 N-glycosylated proteins and 479 N-glycosylation sites were identified from 2 uL human serum samples.This indicates that functionalized mesoporous silica gel has great potential for sample pretreatment in glycoprotein cytology.2.Silicon-based hybrid polymer:Polyhedral oligomeric silsesquioxane?POSS?is a special kind of silicon-based material with stable cage structure and good heat resistance.Using AIBN as initiator,the functional monomer GMA and Acryl-POSS were polymerized via seed-swelling polymerization method,and the optimum proportion of monodisperse PGMA-POSSMA-POSS microspheres was explored.
