The main characteristic of diabetes is persistent high blood sugar.It is a metabolic disease and is also the focus of attention in many fields.The incidence rate of type II diabetes,which is the most dangerous,exceeds 90%of the total incidence rate.At present,the clinical treatment of type II diabetes mainly depends on insulin and chemosynthetic hypoglycemic drugs,among which natural products play an important role in the treatment and prevention of type II diabetes due to their multi-target and multi-channel characteristics.Gallic acid and protocatechuic acid are natural compounds derived from plants,which are safe and have good potential of anti-bacterial,anti-tumor and reverse islet resistance.Considering the presence of o-diphenol hydroxyl and carboxyl groups in gallic acid and protocatechuic acid from the view point of molecular structure,they can form metal ion complexes with metal ions,and have no cytotoxicity to normal cells,so they are ideal ligands.Trivalent chromium can synergize and strengthen the effect of insulin,improve the conversion and utilization of glucose in blood,and reduce blood glucose.In this paper,chromium???complexes were synthesized from gallic acid and protocatechuic acid and trivalent chromium ion,and their structures were characterized.The effects of gallic acid and protocatechuic acid on HepG2 cells and insulin resistance were studied.?1?Taking gallic acid as the ligand and Cr3+as the center ion,the effects of reaction time,pH,reaction temperature and feed ratio on the productivity were investigated respectively,among which pH had the greatest influence on the product.When pH was 6,new chromium?III?gallic acid complex[GA-Cr?III?]was synthesized,which was light green.The results of FT-IR,1H-NMR and ICP etc.showed that the main binding sites of gallic acid and chromium???are o-hydroxy and carboxyl.When the pH is 9,the nanoparticles of stable chromium gallic acid complex were prepared.?2?Taking protocatechuic acid as the ligand and Cr3+as the center ion,the effects of feed ratio and pH on the products were investigated respectively.The results showed that the product was a kind of product,and all the products were chromium containing nanospheres under different pH conditions,so the best condition?pH=6?was selected for the study.?3?HepG2 cells were used for insulin resistance experiment.The results showed that the three chromium complexes prepared for the first time in this study had the effect of reversing insulin resistance.With the increase of drug concentration,the cell survival rate gradually increased,showing a certain dose-dependent.The two chromium gallinate complexes can also significantly improve the survival rate of HepG2 cells,providing a candidate compound for the development of new hypoglycemic drugs;the chromium protocatecholate complexes have no significant effect on the survival rate of HepG2 cells.
