Adenosine triphosphate(ATP)is an significant multifunctional compound in organism.It is mainly produced in mitochondria and participates in many physiological and pathological processes.ATP also plays an indispensable role in colitis.Therefore,the establishment of a method to accurately and sensitively detect ATP is of great importance for studying the mechanism of colitis and other inflammation.As a novel family member of organic metal frameworks(MOFs),zeolitic imidazole frameworks(ZIFs)possesses excellent stability and is widely applied in filed of gas storage and separation,catalysis,small molecule delivery and sensing.The fluorescent probes based on ZIF has been developed gradually in recent years.In this paper,we have designed to used ZIF-90 which has biocompatibility and high stability,as an nano carrier and synthesized three kinds of new nano fluorescent probes to detect ATP.We have successfully applied one of them to ATP imaging in living cells and inflammatory site imaging of mice by ATP sensing.Firstly,A rhodamine-based nanofluorescent probe(NIR@ZIF-90)was synthesized successfully by assembling rhodamine based near-infrared dye into ZIF-90 nanomaterials.The fluorescence probe possesses a near-infrared fluorescence emission with a wavelength of 750 nm when treated with ATP.And a 72 times increases has shown when the probe was treated with 5 m M ATP.Furthermore,the probe has the ability of mitochondrial localization in cell imaging,and can be used to monitor the change of ATP content in inflammatory cells in real time.Moreover,the probe also has successfully achieved the imaging of colitis in mice through ATP sensing.Secondly,A semicyanine-based fluorescent probe(Cy OH@ZIF-90)for ATP detection was synthesized successfully by assembling the semicyanine-based near-infrared dye into ZIF-90 nanomaterials.The synthesis method of the probe is convenient,and it can realize the quantitative detection of ATP by fluorescence intensity signal,and the response time is fast.In addition,compared with other nucleotides and biological ions,the selectivity of the probe has for ATP is nice,and the probe can effectively detect ATP in the p H range of 4-8.Finally,a nano fluorescent probe based on naphthalimide dye(Nap@ZIF-90)was synthesized successfully by synthesizing the naphthalimide-based dye and assembling the dye into ZIF-90 nano materials.The synthesis method of the probe is extremely simple.And the probe can detect ATP quantitatively through the change of fluorescence intensity signal,and the response time of the nano probe for ATP is fast.In addition,compared with other nucleotides and biological ions,the selectivity of the nano fluorescent probe for ATP is also wonderful,and the probe could effectively detect ATP under physiological p H conditions.
