Paeonia suffruticosa is a plant of the genus Paeonia ranunculaceae and is a perennial deciduous shrub.Generally,lectins are present in the rhizomes,leaves,seeds and fruits of plants.There have been no reports of lectin extraction from Paeonia suffruticosa.Therefore,lectin was extracted from Paeonia suffruticosa seeds for research.In this study,Paeonia suffruticosa lectin was isolated and purified,and its biological activities such as coagulation activity,sugar specificity,antioxidant activity,bacteriostatic activity,and effect on cells were studied,as well as preparation of fullerene complexes.The aim is to explore the biological activity of this new type of lectin and provide a basis for the further development and utilization of lectin.The main findings obtained are as follows:The Paeonia suffruticosa lectin was broken up,separated and purified by ammonium sulfate fractionation,Cellulose DE-52 and TSKgel 3000PWXL chromatographic columns,and the protein with coagulation activity and a single symmetrical chromatographic peak was separated and purified.Lactose,L+rhamnose inhibited lectin PSL.SDS-PAGE showed that the relative molecular mass of PSL was 12 kDa.Amino acid composition analysis showed that PSL contained 15 amino acids such as glutamic acid,aspartic acid,and glycine.Among them,the content of glutamic acid is the highest,accounting for 19.66%.Bacteriostatic experiments showed that Paeonia suffruticosa lectin could inhibit Shewanella and Bacillus subtilis.The results of Paeonia suffruticosa lectin on cell proliferation inhibition experiments show that Paeonia suffruticosa lectin can promote mitosis of HBL-100?human breast cells?,ES-2 cells?human ovarian clear cell cancer cells?and mouse spleen cells within a certain concentration range.effect.Paeonia suffruticosa lectin on HBL-100?human breast cells?and ES-2 cells?human ovarian clear cell cancer cells?proliferation experiment results show that Paeonia suffruticosa lectin promotes mitosis in HBL-100 cells and ES-2 cells at low concentrations Role.Studies on mouse spleen cell proliferation studies have shown that Paeonia suffruticosa lectin can significantly promote mouse spleen cell proliferation in a low to medium concentration range,and that when synergistic with CAL,its synergistic effect on mouse spleen cell proliferation is higher than using peony lectin alone.In the study of the preparation of Paeonia suffruticosa lectin and fullerene complexes,Fourier transform infrared spectroscopy analysis revealed that Paeonia suffruticosa lectin can interact with fullerene,and Paeonia suffruticosa lectin was modified by the electrophilic addition reaction between amino and fullerene.To the surface of the carbon cage,multiple hydroxyl groups are also bonded to the fullerene surface.Through antioxidant experiments,it was found that the purified Paeonia suffruticosa lectin and Paeonia suffruticosa lectin fullerene complex have the effects of scavenging DPPH,ABTS,hydroxyl radicals,and superoxide anions.The overall antioxidant activity of the fullerene-lectin complex Better than single Paeonia suffruticosa lectin.
