Astaxanthin is considered as the most powerful antioxidant in the nature,which has bright applications in pharmaceuticals,cosmetics and food because of its properties such as anti-tumor,anti-aging.It is expensive due to limitation of biological source and difficulty of extraction.Lactobacillus is a member of the probiotic family.L.plantarum CHU-R has high value,because it produces astaxanthin and other carotenoids with high safety.In this paper,L.plantarum CHU-R was used as a new biological source of astaxanthin.The cell disruption and supercritical CO2 extraction process was invented under the ethanol system,parameter optimization and industrial enlargement were carried out.?1?The effects of rotation speed?Rs?,ratio of bacteria to beads?A?,ratio of alcohol to bacteria?B?,time?C?,beads?D?on cell disruption?Cd?were studied.Results showed that cell disruption rate exceeds 98%under the condition of Rs=650 r/min,A=0.5,B=0.8,C=4 h,D=different beads.?2?The effects of pressure?P?,temperature?T?,time?t?,granulation ratio?R?,ratio of loading to volume?L?,water content?H?on extraction were studied.The total extraction rate is 12.82%under the conditions of P=20 MPa,T=55 oC,t=80 min,R=0.22,L=0.8,H=30%which is more than 4 times higher than the solvent method.?3?The effects of time,alkali concentration?Ac?and temperature on saponification were studied.Concentration of free astaxanthin reached29.34?g/m L under the conditions of t=9 h,Ac=0.4 mol/L,T=4 oC.Total carotenoid content is20.5 mg/g oil,astaxanthin content is 1.6 mg/g oil with SCCO2.?4?TLC and column chromatography were used to purify the extract,then the main components were identified.The extract contains astaxanthin,?-carotene and other carotenoid,in addition,ergosterol and ten kinds of fatty acids have been found.?5?The effects of solvents,temperature,light,oxygen,etc.on the stability of total carotenoids in extract were studied.The pigment was dissolved in vegetable oil and stored at 4 oC in the dark for 30 days,residual rate was 92.2%.?6?Pilot tests of cell disruption and extraction were conducted,production plan and industrialization analysis were carried out.The results are helpful for the production of astaxanthin.
