Objective: Optimized the background fluorescence quenched-competitive immunochromatographic detection system after set up and applied it to establish a quantitative detection method of single drug dissolved in artificial saliva,like amphetamine,methylenedioxymethamphetamine,and methadone and a simultaneous quantitative detection method of multiple drugs dissolved in artificial saliva,like cocaine,methamphetamine and ketamine.Methods: First,the appropriate amount of anti-drug monoclonal antibody is labeled on colloidal gold by electrostatic adsorption to prepare a gold-antibody conjugate probe,and then the appropriate amount of drug-BSA conjugate and goat anti-mouse Ig G are sequentially coated on the test line and control line of the test strip.Combined with the equipped full-automatic dry-type fluorescence immunoanalyzer,a quantitative detection method for single or multiple drugs targets in artificial saliva was established,and the method was verified later.Results:1.When the diameter of colloidal gold is 21 nm was more suitable for building background fluorescence quenched-competitive immunochromatographic detection system.2.Based on the established system and optimized test conditions can achieve the aim of detecting amphetamine ranged from 0 to 400 ng/m L,methylenedioxymethamphetamine ranged from 0 to 25 ng/m L,and methadone ranged from 0 to 500 ng/m L,all dissolved in artificial saliva.3.Based on the established system and optimized test conditions can achieve the aim of simultaneously detecting cocaine ranged from 0 to 200 ng/m L,methamphetamine ranged from 0 to 40 ng/m L,and ketamine ranged from 0 to 40 ng/m L,all dissolved in artificial saliva.Conclusion: The background fluorescence quenched-competitive immunochromatographic detection system combined chromatographic technology with brilliant separation function and immunoassay technology with high sensitivity and high specificity.Through fiber optic sensing spectrum analysis technology,the rapid andaccurate quantitative detection of multiple drugs breaks the limitation of traditional immunochromatographic assay which performed qualitative identification or semi-quantitative analysis of drugs only,and is expected to become the basis for court statements.
