Ginsenoside Rg5 is a secondary rare ginsenoside obtained by structural modification of protopanaxadiol saponins(PPD).It is a potential natural medicine with good anti-tumor,improving lung inflammation,inhibiting platelet aggregation,improving memory and other pharmacological activities.In this paper,a method of drawing a standard curve of ginsenosides by weighing method is established,and inorganic strong acid salt is used as a catalyst,and ginsenoside Rg5 is efficiently prepared by selective structural modification of protopanaxadiol saponins.Different concentrations of ginsenoside standard solutions are prepared by weighing method,and analyzed by liquid chromatography.The standard curve of ginsenoside was drawn with the concentration as the abscissa and the peak area as the ordinate..The experimental results show that when the standard curve is made by the weighing method,the fitting degree is high,and the R~2 values are all 0.9999 or above The method of preparing the standard curve by the weighing method does not have too high requirements for the pipette and the operator,and has the advantages of simple operation,good accuracy,and the standard solution can be used for a long time(under the condition of no deterioration),which is a practical very strong method for preparing standard curve.The ginsenoside Rg5 was prepared with PPD saponin as raw material and hydrochloric acid as catalyst,and the preparation process was optimized through single factor experiment.The experimental results show that when the concentration of PPD saponin is 20 mg/mL,the reaction solvent is 95%ethanol,the concentration of hydrochloric acid is 0.01 mol/L,the reaction temperature is 70℃,and the reaction time is 3 h,the yield of ginsenoside Rg5 can reach 52.32%.The ginsenoside Rg5 was efficiently prepared with PPD saponin as raw material,high-viscosity chitosan as adsorbent and hydrochloric acid as catalyst,and the preparation process was optimized by single factor experiment and response surface method.The experimental results show that when the solvent is methanol,the concentration of hydrochloric acid is 0.17 mol/L,the amount of high-viscosity chitosan is 20 mg,the reaction temperature is 65℃,and the reaction time is 100 min,the yield of ginsenoside Rg5 can reach 59.49%.The mechanism research shows that high-viscosity chitosan adsorbs PPD saponins through hydrogen bonding in the presence of hydrochloric acid and makes it structurally modified on the surface of chitosan,thereby inhibiting the massive decomposition of ginsenoside Rg5 and improving ginseng Yield of saponin Rg5.In this paper,the yield and preparation efficiency of ginsenoside Rg5 have been greatly improved,and the preparation cost has been effectively reduced by optimizing the process conditions for the preparation of ginsenoside Rg5 catalyzed by hydrochloric acid.The results of this study can provide technical and theoretical support for the preparation of ginsenoside Rg5,and lay a raw material foundation for the research of ginsenoside Rg5 in medicine.
