The reference material has one or more uniform characteristic values, and its characteristic values have various properties such as stability, homogeneity, and transferability.reference material can be used for testing instruments, and can also be used to test the measurement capabilities of laboratory personnel or laboratories. With the introduction of large-scale testing instruments in the laboratory, the demand for reference materials has become larger, and the development of reference materials has become more and more important. In this paper, the mass balance method was used to quantity fipronil, maleic acid and bisphenol A,and a new method for the analysis of human insulin was developed. The research work mainly includes the following aspects:1.The HPLC detection conditions for fipronil,fipronil-desulfinyl,fipronil sulfone were determined.By measuring the organic purity of fipronil and its three derivatives, the moisture contained therein, volatile solvent residues, and non-volatile solid impurities, the results of the mass balance of fipronil and its three derivatives are obtained. It is of great significance to the detection of fipronil pesticide residues in agricultural products and application for primary standard substances for fipronil and its three derivatives.2.The HPLC detection conditions for maleic acid were determined and high purity maleic acid was obtained. The organic purity of maleic acid was accurately obtained.By measuring the moisture content of maleic acid, residual volatile solvents and non-volatile impurities, the purity of the maleic acid by mass balance method was calculated to be 99.88%,which is of great significance for the development of maleic acid standard materials.3.The optimized HPLC detection conditions for bisphenol A(intercomparion number CCQM-K148)samples were determined, and two impurities in the CCQM-K148 sample were qualified and quantified. The purity of CCQM-K148 bisphenol A was determined to be 99.43%by mass balance method. The accurately determined value of bisphenol A and the qualitative and quantitative determination of impurities in it are of great significance for demonstrating the quantify ability of China.4.The HPLC conditions for insulin were optimized, and insulin was purified under the optimized conditions. Then the purified insulin sample was determined by quantitative nuclear magnetic resonance(qNMR)method.Using the purified insulin as an external standard, the commercially available insulin was determined by HPLC and the uncertainty was calculated. The uncertainty of this method for insulin was much lower than the traditional method.A new method is provided for the value assignment of macromolecular organic materials.
