Salvia miltiorrhiza is a traditional Chinese herbal medicine derived from the dry roots and rhizomes of the Salvia miltiorrhiza plant.The chemical composition of Salvia miltiorrhiza are m ainly water-soluble components and fat-soluble components.Among them,the fat-soluble component is mainly a tanshinone-type diterpenoid compound.Tanshinones have extremely high medicinal value,and their development is of great significance to the pharmaceutical and chemical fields.Traditional separation and purification methods for tanshinones have many problems that limit the development of the tanshinone industry.In order to solve the problems existing in the separation and purification process of tanshinones,this topic applies the new separation technology-Membrane Separation Technology to the purification process of tanshinones,and improves the processing amount of tanshinone extract and the purity of tanshinone.The specific work is as follows:(1)Through the screening,the optimal membrane preparation conditions of the ultrafiltration membrane are as follows:the polyethersulfone concentration is 22wt.%,the feeler gauge thickness is 0.15mm,and the concentration of polyethylene glycol and glycerol additive is 2wt.%.The pure water flux of the polyethersulfone ultrafiltration membrane prepared at this time was 155 L/m2h,and the rejection rate of bovine serum albumin reached 82%.Ultrafiltration membranes were prepared by using different polyethersulfone concentrations of casting solution,and applied to the preliminary step-by-step impurity removal treatment oftanshinone extract.The purity of tanshinone increased from 14.3%to 16.7%.(2)The optimal membrane preparation conditions for nanofiltration membranes by single factor control method are:oil phase monomer TMC concentration 0.5wt.%,water phase monomer PIP concentration 1.0wt.%,pH of aqueous phase monomer solution is 9.5,the water phase monomer treatment time 10 min,the interfacial polymerization reaction time is 60s.At this time,the pure water flux of the nanofiltration membrane is 58L/m2hMPa,the rejection rate for sodium ions is about 30%,and the retention rate of magnesium ions can reach more than 90%.By applying different nanofiltration membranes to the purification and concentration process of tanshinone,the treatment amount is greatly improved,and the purity of tanshinone purified by nanofiltration membrane can reach 51%.
