Teicoplanin(TE)and vancomycin(VCM)were the most commonly used to treatment of severe Gram-positive infections in the clinical,commonly,that blood concentration has a significant impact on treatment.TE was a concentration-dependent antibiotic,which's blood concentration affected inhibitory effect for bacteria,directly.The adverse reactions of VCM,such as ototoxicity,nephrotoxicity,would occur with the increase of VCM concentration.Molecularly imprinted membrane(MIM)was prepared by TE and VCM as templates,respectively,which can be used to detect the concentration of TE and VCM in biological samples.In the part of TE MIM,molecularly imprinted membrane(MIM)with TE as a template and other related different material membranes(organic nylon microporous membranes(NN),polyvinylidene fluoride(PVDF)membranes,polypropylene membranes)as a support respectively were synthesized for the selective absorption of TE,which can be used for pretreatment biological samples and rapid determination of TE.The results showed that PVDF MIM had the best selectivity of the other three synthesized membranes to be used in the work.And the results of static adsorption experiment and kinetic adsorption experiment showed that the maximum loading of TE PVDF MIM was 85ug/cm~2 and the optimum adsorption time was 90 min.10%acetic acid methanol solution(v:v)was used as the eluent,which's UV standard curve of Y=0.0093X+0.0131 and R~2=0.9995 was plotted.In the presence of VCM which had similar structure whith TE,TE PVDF MIM also showed selective adsorption capacity for TE.After the desorption experiment,the optimal desorption time was 30 min.The recovery both in pure solution or serum was good by using this method.And also the precision,stability,and repeatability of this method were investigated at the same time.In the part of VCM MIM,the MIM with VCM as a template and other related different material membranes(organic nylon microporous membranes,PVDF membranes,polypropylene membranes)as a support respectively were synthesized for the selective absorption of VCM,which can be used for pretreatment biological samples and rapid determination of VCM.The optimal ratio of template and functional monomer was 1:4 which was simulate by Gaussian.The imprinting layer was successfully synthesized on both VCM NN MIM and VCM PVDF MIM by adsorption experiment and scanning electron microscopy(SEM).And the VCM PVDF MIM was selected for subsequent experiment,which had better adsorption effect for VCM than NN MIM.And the results of static adsorption experiment and kinetic adsorption experiment showed that the maximum loading of TE PVDF MIM was 23.5ug/cm~2 and the optimum adsorption time was 120 min.And the VCM PVDF non-molecularly imprinted membrane(NIM)had no absorption to VCM.In addition,in the presence of TE which had similar structure whith VCM,VCM PVDF MIM also showed selective adsorption capacity for VCM.Then,the eluent was screened,and0,1 mol/L KOH aqueous solution and methanol volume ratio of 1:4 has the best elution effect,which's UV standard curve was Y=0.0311X+0.0724 R~2=0.9990.After the desorption experiment,the optimal desorption time was 20 min.The recovery both in pure solution or serum has good effect of VCM by using this method.And also the precision,stability,and repeatability of this method were investigated at the same time.
