Tieguanyin tea is rich in tea polyphenols,theanine,caffeine and other active ingredients,it has many effective health benefits,such as the function of reducing blood sugar and fats,anti-oxidation,anti-tumor,etc.The potential efficacy mechanism of those functions may related to anti-inflammation.There are many researches on the anti-inflammatory effects of active ingredients of tea in domestic and foreign countries,but little research on the anti-inflammatory function of tea extract mixtures.In this research,we test chemical compositions,sensory evaluations,and anti-inflammation fuction in vivo and in vitro of eight different flavor types and years of Tieguanyin tea samples,compare and analyze the influence of different flavors and storage years on chemical composition and sensory quality of Tieguanyin,and study the antiinflammatory activity and its possible mechanism.The results of the study are as follows:Firstly,tested the composition and evaluated the sensory of 8 different fragrances or storage years of Anxi Tieguanyin,and analyzed the correlation between sensory quality and chemical composition.The results shown that the difference between sensory quality of Nong Xiang Tieguanyin and Qing Xiang Tieguanyin is mainly because different manufacturing processes made of different ingredients.With the increase of storage year,the moisture content and gallic acid content of Chenxiang Tieguanyin showed an upward trend,and the contents of tea polyphenols,catechins,amino acids,soluble sugars are fluctuating.The water content was significantly negatively correlated with tea quality scores(p<0.05),and tea polyphenols,catechins,total amino acids,theanine,soluble sugars and were significantly positively correlated with tea quality scores(p<0.05).Secondly,models of auricular swelling induced by xylene and acetic acid induced experimental peritonitis in mice was established to study the anti-inflammatory effects of 8 Anxi Tieguanyin tea extracts.The results shown Tieguanyin tea extract has certain improvement effect on acute exudative inflammation: 8 Tieguanyin tea extracts had certain inhibitory effect on xylene induced auricle swelling and the increase of peritoneal capillary permeability induced by acetic acid,but the inhibition were different.Nong Xiang Tieguanyin,Qing Xiang Tieguanyin have significant inhibitory effect(p<0.01),and the effect is positively correlated with dose.The anti-inflammatory activity of tea in this year was stronger than aged tea.Thirdly,used LPS to induce RAW64.7 cells to establish an inflammatory response model.Detected the secretion of NO and IL-6 and the expression of iNOS,COX-2,IL-6,TNF-? and MCP-1 mRNA to investigate the inhibitory effect of the Nong Xiang Tieguanyin tea extract in vitro.The results shown Tieguanyin tea extract can significantly inhibit the release of NO and IL-6 induced by LPS in RAW264.7 cells.The inhibition effect was via down regulating the gene expression of iNOS and COX-2 and the gene expression of proinflammatory cytokines IL-6,TNF-? and MCP-1.The inhibition effect was significant at high concentration(p<0.01).Finally,Detected key proteins of NF-?B,an inflammatory signaling pathway,by Wstern blot,to investigate the possible mechanism of anti-inflammatory effect of Tieguanyin tea extract in RAW264.7 cells.The results shown Tieguanyin tea extract can inhibit the activation of NF-?B signaling pathway,inhibit the phosphorylation of IKK?(11)I?B and p65,reduce the NF-?B p65 binding to target gene so that the release of various inflammatory factors were suppressed.The anti-inflammatory activity of high concentration of tea extract is better than 20 ?g/mL indomethacin(p<0.01).
