The medicinal values of Angelica keiskei are being paid attention for the effects of anti-aging,anti-ulcer,anti-thrombosis as well as balancing blood pressure,lipids and blood sugar,etc..Polysaccharide of Angelica keiskeias the water soluble polysaccharide possesses great medicinal and edible values.The Response Analysis methodology is being applied for this research to obtain the optimized parameters of water extraction for polysaccharide which are 65? extraction temperature,2.4h extraction time and the ratio of1:16for ingredient/water.A yield rate of 26.37% was calculated under the extraction process with the optimized parameters.F25 and F40 were obtained after the ethanol precipitation of neutral polysaccharide.After extraction of Angelica keiskei polysaccharides,two kinds of polysaccharides which are acidic polysaccharide and neutral polysaccharide were obtained under the process of CTAB precipitation and dialysis separation/purification.The yield rates are 23.22% and 62.01%,respectively.F25 and F40 were obtained as the reaction substances after the ethanol precipitation of neutral polysaccharide to result the yield rates of 33 and 26,respectively.The UV spectrums showed that there wereno nucleic acid and protein in therfactions of Angelica keiskei polysaccharide.All the fractions of Angelica keiskei polysaccharide contained specific polysaccharide absorption peaks as well as Type D furan ring and C-H bonds when scanned by IR.Both netural polysaccharide and F25 contained ?-Glycoside bond.The GPC analysis results of fractions of Angelica keiskei polysaccharide showed the heterogeneous molecular weight of neutral polysaccharide and F25 and homogenous molecular weight of F40,which is187.36 kDa and 57.69 kDa.Rham,Man,Glu,Ara and Gal were eluted out from neutral polysaccharide by HPLC with mole ratios of 1.00:1.96:2.75:3.21:11.39.The F25 and F40 mainly contained Rham,Glu,Ara and Gal with the ratios of 1.00:2.44:5.74:8.32 and 1.00:0.98:1.33:1.92,respectively.The primary structure may beAG.
