Drug nano-carriers can significantly change the delivery and distribution of drug in vivo.Especially the targeting nano-carriers can increase the concentration of drug at the targeting site,so they can improve the efficacy at the same time reduce the toxicity.The aim of this study is to provide the basis for the design of targeting nano-carriers by in-depth analysis the delivery and targeting/off-target effect of drug nano-carriers through in vivo and in vitro models.Firstly,we choose three liposomes with different surface chemical properties to establish the detection method for evaluating the interaction between nano-carriers and cell,protein.And we apply the method to analyze the different delivery process of those liposomes,especially the interaction with serum protein and white blood cells.Secondly,we establish a series of analysis and characterization methods to analyze the targeting liposomes’ targeting/off-target effect in different tissues.Finally,on the basis of the study mentioned above,we design and optimize the gene carriers based on DOTAP cationic lipid to target monocyte.In vitro cell experiments,it has been proved to have stable and great transfection even in serum.Through the study of this subject,we establish a new method to evaluate the interaction process and targeting effect of nano-carriers in vitro and in vivo,meanwhile provide a basis for the optimization and application of nano-carriers.
