Malachite Green(MG)is a synthetic organic material. The chemical property of malachite green is triphenylmethane.Colored malachite green is used as a dyeing agent in the industry, and it can kill bacteria, kill fungi, kill parasites. In aquaculture, many fishermen use it to prevent infectious diseases of various fish species. The long-term consumption of human can cause cancer, and many countries have enacted laws prohibiting the addition of malachite green in food. Therefore,China’s restrictions on the import of aquatic products have become increasingly stringent,so rapid detection of agricultural products is a promising application. Many studies have shown that Malachite green enters the aquatic system and enters the circulatory system. The aquatic animals eventually metabolize into fat-soluble Leuco Malachite Green(LMG).This article describes the production, toxicity, metabolism and detection methods of malachite green in aquatic products, and proposes methods for the residual toxicity of malachite green. The substance is mainly used to distinguish the material of Malachite Green and Malachite Green’s metabolite leucomalachite green.Because of the different molecular structure of each kind of material, the resonance frequency between atoms is also different, and the specific spectrum generated is the only thing that can determine the material molecules. When the material is excited, the excitation radiation generated is not only a kind of fluorescence but also Raman scattering. The wavelength of Raman scattering is constant under certain incident excitation light.Absorption spectrum and emission spectrum are relative, absorption spectrum can also be used to detect substances.Simultaneous measurement using fluorescence and UV spectrophotometry can provide an effective persuasive power to prove a substance. This article aims to explore a rapid and effective means to identify malachite green and to know its detection limit. The experiment uses fluorescence detection method to analyze the content of malachite green. The experimental results show that a single light source with an excitation wavelength of 265 nm can be used to determine the residual amount of leucomalachite green at the emission wavelength of 360 nm.The linear correlation is good in the range of 0.01 ppm to 6 ppm, satisfying Beer-Lambert’s law,and the correlation coefficient is 95.73.% and 97.79%,with a minimum detection limit of 0.09 ppm.
