In order to explore the screening and optimization of toxicology,health care function,hygiene and production technology rats and mice were taken as the research objects to test its safety in toxicology and health care function.The contents of heavy metals and microorganisms in the product were detected in hygienic tests.The production process is optimized by comparing and optimizing the ethanol extraction and water extraction.The results are as follows:Product is made of seabuckthorn,astragalus,ginseng and Paecilomyces bat moth powder as raw materials.Ginseng and seabuckthorn are extracted by alcohol.The optimum technological conditions of alcohol extraction of this product are ginseng,Seabuckthorn and75% ethanol reflux extraction twice,each time adding 7 times of solvent,each reflux for 1.5hours.The best technological conditions for water extraction of this product were determined as ginseng,Seabuckthorn and Astragalus membranaceus,which were decocted twice with 8times of water for 1.5 hours each time.It also includes separation,concentration,drying,crushing,shaping,capsule filling and polishing,packaging and inspection.The acute oral MTD was more than 15g/kg.bw in rats,it shows that it is non-toxic.Three genotoxicity tests of bone marrow micronucleus test,sperm malformation test and Ames test in mice were all negative..No abnormal changes in animal health,biochemical,hematological parameters and organ morphology were observed in the 30-day feeding test of rats.Based on this,it was preliminarily estimated that the maximum non-effective dose of the product was more than 4.0g/kg.bw(That is more than 100 times the recommended intake of human body.).After 30 days of oral administration of 0.4g/kg.bw,0.8g/kg.bw and 1.2g/kg.bw doses to mice(Balb/c)respectively,no adverse effects on weight and weight of mice were observed.In the determination of organ/body weight ratio of mice,there was no significant difference in organ/body weight ratio between each dose group mice and control group mice(P > 0.05).In the delayed allergic reaction(toe swelling)test of mice,there was a significant difference in the degree of swelling of the soles between the 0.4g/kg.bw and 0.8g/kg.bw groups compared with the control group(P < 0.01).In the mouse spleen lymphocyte transformation test(MTT method),the difference of optical density between 0.8g/kg.bw dose group and control group was significant(P<0.05).In the experiment of phagocytosis of chicken erythrocyte by mouse peritoneal macrophages,the percentage and index of phagocytosis of chicken erythrocyte by L2g/kg.bw group were significantly different from those of control group(P < 0.05)and very significant difference(P < 0.01).In the NK cell activity test of mice,the NK cell activity rate of each dose group was significantly different from that of control group(P < 0.01).According to the evaluation criteria,it can be concluded that the test has the function of enhancing immunity.For three months,the product met the standard in sensory indexes such as color,odor,etc.The total efficacy/marker components such as total saponins greater than 1 g/100 g,adenosine content greater than 130 mg/100 g,physical and chemical indexes such as arsenic < 0.10mg/kg,lead < 0.20 mg/kg,mercury < 0.05 mg/kg,HCH < 0.01 mg/kg,DDT < 0.02 mg/kg,net content and negative deviation of bacterial colonies in microbial indexes.No Staphylococcus aureus,Salmonella,Shigella and Streptococcus haemolyticus were detected.In conclusion,the product meets the standard in hygienic testing.Through the research on toxicological test,health function test,hygienic test and production technology,the whole research and production process of a kind of health care product were systematically elaborated,which provided some reference for the research of other health care products.
