For heavy physical workers,especially those who have been in emergency environment for a long time,are prone to symptoms such as anxiety,forgetfulness,and depression.Many of the active ingredients of natural products have the effect of improving the body’s fatigue.Protein from Pinus Koraiensis Sieb.et.ZUCC.,blueberry anthocyanins and lycium polysaccharides have become the focus research in recent years due to their unique physical and chemical properties and physiological functions.In this study,the nutrition calculator was used to assist in formula screening.The Pinus Koraiensis Sieb.et.ZUCC were used as raw materials,supplemented with blueberry anthocyanins and lycium polysaccharides.The formulations of the prepared composite preparations were tested by single factor test,response surface optimization experiment and orthogonal optimization test.the anti-fatigue effect was evaluated by establishing a mouse fatigue swimming model.The formulation base on the Protein composite anti-fatigue preparation was screened and optimized.Composite anti-fatigue liquid and solid preparations were prepared by rotary evaporation concentration and three-dimensional granulation.The single factor test was carried out with the ratio of raw and auxiliary materials,the proportion of auxiliary materials,the amount of sucrose,the solid-liquid ratio and the amount of citric acid.The ratio of raw materials and materials,the solid-liquid ratio,the amount of citric acid and the amount of sucrose were determined.The amount of addition was the main sensory influence factor.The orthogonal test was carried out with L9(34).The best formula was obtained by sensory evaluation,that is,the sucrose addition amount was 18 g,the citric acid addition amount was 0.08 g,and the original auxiliary material ratio was 3:1.The solid-liquid ratio is 1:8,and the blueberry mass ratio is 1:3.To study the rheological properties of the composite anti-fatigue preparation and the effect of powder properties on stability.The best combinations of stabilizers was obtained by single factor screening and response surface analysis.The gelatin content was 0.2 mg/mL,the xanthan gum content was 0.1 mg/mL,and the Carboxymethylcellulose sodium content was 0.05 mg/mL.The optimal combination of emulsifiers was single.The content of glycolipid was 0.2 mg/mL,the content of sucrose ester was 0.1 mg/mL,and the content of Tween-80 was 0.05 mg/mL.According to the measurement results of apparent viscosity,shear rate-shear stress curve and rheological parameters at the corresponding concentration,the stability of liquid preparations under different dilution concentrations was analyzed.The results showed that the composite liquid preparations at different dilution concentrations showed performance.The pseudoplastic flow behavior of a typical non-Newtonian fluid whose solution viscosity decreases with increasing shear rate.The internal structure analysis of the solid preparation was carried out by particle size analyzer and SEM scanning electron microscope.The powder properties of the solid preparation were comprehensively evaluated by analyzing the solubility,dispersibility,angle of repose and Karjner index.The results showed that the particle size distribution of the Protein from Pinus Koraiensis Sieb.et.ZUCC.composite anti-fatigue solid preparation was uniform and the stability was good.Physical and chemical analysis of anti-fatigue of Protein complex was carried out and a mouse fatigue swimming model was established.The anti-fatigue effect of the composite preparation was studied by behavioral test and biochemical index determination.The results showed that the soluble solid content of the Protein composite anti-fatigue preparation was 8.67%,the pH value was acidic,the soluble protein content was 15.3%,and the moisture content was 68%.The microbial indicators were in line with national standards.The animal model experiments showed that the composite preparation did not affect the body weight change and normal growth of the mice.The preparation of the preparation can significantly prolong the time of weight-bearing swimming and exhaustion,increase the organ index,and effectively improve blood sugar and muscle glycogen.The content of hepatic glycogen reduces the content of lactic acid and serum urea nitrogen in the blood.In addition,it can significantly improve the density distribution of elastic fibers and collagen fibers in the skin of mice,and has a certain protective effect on fatigue-damaged skin.Fatigue stress response is common in the work of heavy manual workers.Therefore,the research of this subject has high application value and guiding significance for further development of natural products as anti-fatigue special function protection products.
