The in vivo fate of fat nanoemulsion injections and propofol fat nanoemulsion injections were tracked by imaging tools and HPLC method simultaneously.In this study,a highly sensitive near-infrared environment-responsive fluorescent probe(P2)was used to label nanoemulsions.This probe can be employed to identify the intact nanoparticles in vivo due to strong aggregate caused quenching(ACQ)effect.At the same time,a selective and sensitive HPLC method for the analysis of propofol in biological samples was developed.Analysis of blood-borne fluorescence reveals rapid clearance of nanoparticles,which shows ‘‘L''-type blood pharmacokinetic profiles.Following intravenous administration,the nanoparticles concomitantly accumulate in RES organs such as liver,spleen and lung,and are cleared from body circulation mostly after 12 h.However,propofol is more widely distributed than nanoparticles and absolutely cleared in 1 h.The drug molecular can even cross blood-brain barrier(BBB)and blood-testosterone barrier(BTB).These intuitional results are of significance in understanding the in vivo fate of nanoparticles and packaged drug,which can inspire deep thinking of this updated concept.
