Biothiols such as cysteine(Cys),homocysteine(Hcy),and glutathione(GSH),which play important roles in biological systems.Abnormal biothiols can cause a series of related diseases in vivo.There are many methods to detect biothiols,such as high performance liquid chromatography(HPLC),electrochemical method,electrophoresis,mass analysis and so on.However,these methods have some limitations due to cumbersome operation and high cost.Therefore,it is very important to design probes for detection of biothiol.Many probes have been reported to detect biothiols,but they are difficult to distinguish because of their similar structures.Therefore,it is of vital importance to detect and discriminate Cys/Hcy and GSH.The first chapter mainly introduces the importance of biothiols and the current detection methods of biothiols.This paper introduces the basic properties and applications of coumarin,briefly describes the basic properties and applications of NBD,summarizes the basic properties and applications of diarylene,and finally puts forward the research content of this paper.In the second chapter,coumarin and NBD were used to synthesize the probe 1.The probe can achieve the effect of naked eye detection of biothiol.At the excitation wavelength of 330 nm,the probe could detect Cys/Hcy and GSH from other amino acids.When the excitation was 460 nm,the probe could detect Cys/Hcy from GSH.In addition,the probe was successfully used to detect thiols in living Hela cells with low cytotoxicity.In chapter 3,coumarin and p-nitrophenylthiophenol were used to synthesize probe 2,which could efficiently detect Cys/Hcy and GSH from other amino acids.After the addition of Cys/Hcy and GSH repectevily,probe 2 showed obvious absorption peaks at 496 nm,corresponding to a color change from colorless to pale yellow.In addition,the probe can be used in logic gates.In chapter 4,according to the photochromic properties of diarylethylene,probe3 containing diarylethylene derivatives are synthesized.Probe 3 exhibited goodphotochromic properties.Secondly,probe 3 can detect biothiols with the naked eye from other amino acids.After the addition of Cys,Hcy and GSH,probe 3 solution changed from colorless to light orange,light yellow and light yellow,respectively.The color of the solution changed to dark pink,light green,and light green upon.When the excitation was 470 nm,probe 3 was able to detect Cys and Hcy.A significant fluorescence emission peak appeared at 570 nm,corresponding to a color change from black to yellow-green.In addition,probe 3 can be used in logic gates.
