Fried food in our country has a rich variety and edible history,such as chips,biscuits,coffee,toast,instant noodles and other hot processed food in the processing process will produce acrylamide.With the continuous development of food detection technology and medicine,acrylamide was widely concerned in 2002,and its neurotoxic and carcinogenic effects were mainly reported.Liquid chromatography tandem mass spectrometry(LC-MS/MS)and gas chromatography tandem mass spectrometry(GC-MS)are the current standard detection methods,with high sensitivity,stability,reproducibility and selectivity.The only deficiency is that the test cost is too high,and the experimentors need to be very skilled in the operation,so they cannot meet the routine detection of acrylamide.At present,the rapid detection methods include elisa,computer vision,electrochemical biosensor and fluorescence detection.This paper mainly focuses on the fluorescence detection method to try three detection methods,respectively gas chromatography connected with hydrogen flame ionization detector(GC-FID)method,high performance liquid chromatography connected with fluorescence detector(HPLC-FLD)method,high performance liquid chromatography connected with laser induced fluorescence detector(HPLC-LIF)method.It was found in the experiment that the detection limit of GC-FID was relatively low,which could only reach the level of 10-4mol/L.However,the experiment in this chapter was mainly to verify the extraction method of acrylamide,so as to provide support for subsequent experiments.The high performance liquid chromatograph is connected to the fluorescence detector method and the high performance liquid chromatograph is connected to the laser induced fluorescence detector method.The fluorescence reagent is used as the marker.The single factor optimization of the reaction conditions in the test is also the top priority,optimal acidic alumina catalyst is 1.0 g,the best reaction temperature is 75 0C,the best reaction time is 6 h,excessive rate is 3 times the best dye.The correlation coefficient of The high performance liquid chromatograph is connected to the fluorescence detector method standard curve was 0.9982,the recovery was 99.3%,the recovery relative standard deviation was 1.64%,and the minimum detection limit was 3.38×10-8 mol/L.The correlation coefficient of high performance liquid chromatography connected with laser induced fluorescence detector(HPLC-LIF)method standard curve was 0,9988,the recovery was 100.9%,the recovery relative standard deviation was 1.81%,and the minimum detection limit was3.52×10-9 mol/L.Based on the fluorescence characteristics of dyes,acrylamide extracted was reacted with the two methods to form products containing fluorescent groups and then the content of acrylamide in the sample was determined.Compared with the mainstream detection methods,this method is simple?easy?saves experiment cost and has practical application value.
