China,known as the "kingdom of bamboo",is the country with the most bamboo in the world.Anhui province is rich in bamboo resources,covering 200,000 hectares.In addition,there are nearly 12 genera and 72 species.Moso bamboo leave is an important forestry resource with large capacity,moderate price and excellent efficiency,which is as well as Ginkgo biloba.Recently,extraction and utilization of active ingredients is a hot topic in bamboo leaves filed.However,it is rare that application researches of active ingredients from bamboo leaves in daily necessities.Along with the society,people’s love for pure natural products is growing,while the requirement for commodity is getting higher and higher.Therefore,the development of bamboo leaf extract to a new type of natural antibacterial and bactericidal material not only has important application value but also has strong innovation.In this project,the widely distributed moso bamboo leaves were selected as objects,water and ethanol as extraction solvent,it is optimized the extraction process of active ingredients by vibration and ultrasonic technology combination of liquid chromatography and mass spectrometry method.Additionally,bacteriostasis experiment was conducted by extraction from different solvent.To identify the bacteriostasis component,gas chromatography-mass spectrometry(GC-MS),liquid chromatography-mass spectrometry(UPLC-MS)were used to separate and analyze the structure of the extraction.Furthermore,the extraction was added to the soap formula to explore its application in the antibacterial field.This project will provide basic data and scientific basis for further exploitation of bamboo resources.The concrete research content is as follows:Moso bamboo leaf and branch were selected as objects,water and 80%ethanol-water as extraction solvents,by calculation the peak area in total ion chromatogram and extract ion chromatogram of UPLC-MS,it is optimized the final extraction process.80%ethanol-water was the extraction solvent,solid-liquid ratio was 1:10,the mixture was thermostat shaken for 4h,ultrasound(80Hz)for lh.Bacteriostasis experiment was conducted by extraction from different solvent using paper filtering method.It is showed that the antibacterial activity of bamboo leaf extract was significantly greater than that of shoots;the ethyl acetate extractions showed moderate antibacterial activity against Staphylococcus aureus,Escherichia coli,Salmonella and Bacillus subtilis;80%ethanol-water extraction exhibits moderate against Staphylococcus aureus,Escherichia coli and Salmonella,and only weak against Bacillus subtilis.Moso bamboo leaf,fresh leaf and branch were extracted by 80%ethanol,separately.Using UPLC-MS technology,24,20 and 19 components were detected from the bamboo different parts,11 of them were detected in 3 samples,including vitexin,isovite,valerian,isoorientin et al.By the area comparison method,it is showed that the content of the detected components in bamboo leaves was 4 times than that of bamboo branches.The volatile components of bamboo leaves were separated and identified by GC-MS.A total of 94 compounds were detected by NIST library,including 36 alkanes,20 esters,4 alcohols,6 acids,6 ketones,5 aldehydes,1 ether and 7 kinds of other substances.Furthermore,extraction solvent is special for different compound.Ester is the most in ethyl acetate part;alkane is the maximum in petroleum ether phase;ketone is the richest in 80%ethanol-water initial extract has the most.After extraction process optimization and bacteriostatic activity detection,80%ethanol-water initial extract was concentrated and added in the soap.By comparing commercial and bamboo soap using TA.XT.Plus physical property tester,the hardness,viscosity,elasticity and foam stability were tested.The result showed that the bamboo soap has the characteristics of low hardness,good viscosity elasticity,high foam and excellent stability.At the same time,the antibacterial experiments were carried out on the handmade essential oil soap of phyllostachys pubescens.
