Studies On The Secondary Metabolites Of Two Endophytic Strains Chaetomium Globosum&Daldinia Eschscholzii Fermented In Red Einseng Medium

Abstract/Summary:
Endophytes have complex metabolic pathways and can produce chemical components with rich structure and diverse biological activities.The fermentation of endophytic fungus from medicinal plants can also obtain the pharmacological active ingredients of rare Chinese medicinal materials.So,some problems such as the waste of scarce medicinal plant resources and the limitation of medicinal use can be effectively avoided.In this thesis,in order to obtain more compounds with novel structure and diverse biological activities,red ginseng was used as culture medium to culture endophytic fungi.In the first chapter,the research progress about chemical constituents and biological activities of red ginseng and Chaetomium globosum in the last ten years was summari?zed.The second chapter was mainly about the research on secondary metabolites and their biological activities from Chaetomium globosumJC-H8*1 from Dendrobium nobile Lindl fermented in red ginseng medium.Decoction of red ginseng was selected as culture medium.Combination the traditional separation methods with the high performance liquid chromatography(HPLC)separation technology,17 compounds were separated from the fermented liquid and mycelium.They were identificated by NMR,HR-ESIMS,CD,UV,IR and ORD,including a new natural product cazaldehyde B(13*),a new compound panaxylactone(15*),and 15 known compounds.The structure types of compounds involved cytochalasins,polyacetylene,polyketones,azaphilones,sterols,terpenes and nucleosides.The results of the bioactivity screening of some compounds showed that compound 13*had a strong inhibitory activity for the production of NO at 50μM,with IC50 value 40.12±0.85μM,while the positive control L-NMMAIC50 value was 39.26±0.91μm。In addition,compound 15*was generated on which conditions of fermatation by HPLC.The third chapter was mainly about the the research on secondary metabolites and their biological activities from Daldinia eschscholzii GC-J7*1 from Dendrobium chiysotoxum Lindl fermented in red ginseng medium.Solid fermentation of the Daldinia eschscholzii GC-J7*1 was carried out by red ginseng.15 compounds were obtained from the the EtOAc extract of solid culture of Daldinia eschscholzii GC-J7*1,including 2 new compounds,8-hydroxyl-helicascolidea(7*),daldinsin(8*),and 13 known compounds.Their structures were characterized by NMR spectroscopy and ECD experiments.The structure types of the compounds involved benzopyranes,naphthol,saponins,panaxytriol and so on.The activity screening of new compounds showed that compound 8*had moderate inhibitory activity against acetylcholinesterase(AChE)at the concentration of 50μM,with an inhibitory rate of 38.78%,while the inhibitory rate of the positive control drug,tacrine,was 64.88%.The last chapter,the coculture of two strains of Chaetomium globosum JC-H8*land lshsy-5 from different plant on solid red ginseng medium was carried out.Then 4 compounds were obtained from the the EtOAc extract of solid fermentation of the two strains.panaxytetraol A(1*),panaxytetraol B(2*),panaxydol(3),tetradeca-13-ene-1,3-diyne-6,7-diol(4),including 2 new compounds:panaxytetraol A(1*),panaxytetraol B(2*).Their structures were characterized by NMR spectroscopy and HR-ESIMS.
