Phytic acid is also called inositol hexaphosphate(IP6)and exists widely in plant grain.Phytic acid can chelate minerals,protein and other nutrients with extremely strong electronegativity,thus,it is regarded as antinutritional factors.Phytic acid,as the inorganic phosphorus library,can be degraded by phytase during the germination and release inorganic phosphorus to support plants growth.Lower inositol phosphate was the degradation product of phytic acid,and it has a variety of biological activity and pharmacological effects,such as participate in cell signal transduction,promoting cancer cell apoptosis and preventing diabetes without anti-nutritional effects.Meanwhile,Ins(1,4,5)P3 is the specific product of PI-PLC pathway,which plays a important role in signal transduction.In the present study mung bean sprouts were chosen as experimental materials and phytic acid content,low-level basic content,phytic acid phosphate enzyme and PI-PLC activity and related gene expressions were studied in the mung bean sprouts with different kinds and concentrations of chlorate.As the same time,the effects of combined treatment on biochemical changes and antioxidant ability were studied.The main results are as follows:1.Phytase activity and phytic acid content were studied in mung bean sprouts under KCl,NaCl and CaCl2(2 mmol/L)treatments.The phytase activity and phytic acid content were used to screen the optimal concentrations of chlorizated salts which have function in reducing phytic acid content.The results show that the three kinds of chlorate all had the effect in promoting geminated mung bean growth and NaCl,CaCl2 degraded phytic acid content;1.6 mmol/L NaCl and 6 mmol/L CaCl2 had the best effect on phytic acid degradation.Furthermore,the superposed effect on phytic acid degradation was found with NaCl and CaCl2,the optimal concentrations of NaCl and CaCl2 for joint treatment by response surface analysis were 1.68 mmol/L NaCl and 6.40 mmol/L CaCl2.In this condition,the phytic acid content was reduced to 8.04?g/plant,which reduced by 88.16%.2.Lower inositol phosphate,phytic acid metabolism related enzyme activity and gene expression changes were studied in germinated mung bean with NaCl,CaCl2 and NaCl-CaCl2.The results show that mung bean sprouts with NaCl-CaCl2 contained the lowest phytic acid and highest low-level inositol phosphate.Otherwise,IP3 content increased most with NaCl-CaCl2 treatment,which increased by 103.33%.NaCI,CaCl2 and NaCl-CaCl2 have promoted the process of PI-PLC to produce Ins(1,4,5)P3,and improved the activity of phospholipase C and PI-PLCgene expression.But,NaCl-CaCl2 effect is better than NaCl and CaCl2.3.Phytic acid content,lower inositol phosphate content,PI-PLC activity and related gene expression were measured in mung bean sprouts with NaCl?CaCl2 and NaCl-CaCl2 treatments and combined with neomycin.The results show that effects of PI-PLC pathway and the enrichment of lower inositol phosphate were promoted by CaCl2 and NaCl-CaCl2.Meanwhile,PI-PLC pathway was suppressed by neomycin which can reduce phospholipase activity and PI-PLC gene expression,and the increment of Ins(1,4,5)P3 was mainly produced by PI-PLC pathway.4.Growth performance,endogenous hormone,nutrients,adenine phosphate(ATP,ADP and AMP),the Krebs cycle related enzyme activity(CCO,SDH,Ca2+-ATPase and H+-ATPase),radical scavenging capacity and the change of the activity of antioxidant enzymes were studied in mung bean sprouts with NaCl,CaCl2 and NaCl-CaCl2 treatments,As the results showed,the high level of energy metabolism,growth speed,endogenous plant hormone and nutrient content was found in mung bean sprouts under NaCl-CaCl2.Meanwhile,indoleacetic acid(IAA)and brassinosteroid(BR)contents were improved significantly by NaCl,CaCl2 and NaCl-CaCl2 treatments,at the same time,the OH-· scavenging capacity and autioxidant enzyme activitiy were higher than the control which induced the enhancement total antioxidant capacity.
