Porous materials is a type of material composed of closed or opeded pores.The common porous materials include silica,activated carbon,metal-organic frameworks(MOF),covalent organic frameworks(COF)and so on.In recent years,the research on the functionalization of porous materials has received extensive attention.Photodynamic therapy(PDT)is a new type of cancer treatment technology,which has developed rapidly at the end of the 20th century due to its minimally invasive,non-resistance and other advantages.Phthalocyanine photosensitizers are a class of efficient photosensitizers,but phthalocyanines are prone to aggregation and causing photodynamic activity to decline or even disappear.Due to the high spatial dispersibility and modificability of porous materials,builted porous materials based on phthalocyanine could solve the problems of aggregation and insufficient targeting capacity of the phthalocyanines in some degree.The main work and results of this paper are briefly described as follows:(1)EuBTC is a type of MOFs materials with rare earth metal nodes.Due to its characteristics of both porous material and fluorescence of rare earth metal,EuBTC has attracted much attention in the fields of biology and chemistry.In this paper,the phthalocyanine photosensitizer and the targeting molecule folic acid were covalently attached to the MOF material by post-modification and the targeting molecule was linked to improve its targeting ability.Next,a certain amount of chemotherapy drug doxorubicin was loaded.EuBTC,EuBTC-Pc,FA-EuBTC-Pc,and DOX@FA-EuBTC-Pc were prepared.The synthesized materials were characterized by SEM、XRD、IR and Size and the in vitro photodynamic activity of the synthesized materials was studied too.The results showed that the EuBTC-Pc nanosystem with phthalocyanine had a good photodynamic inactivation ability to tumor cells;due to the introduction of folic acid,FA-EuBTC-Pc nanosystem was well targeted to Hela cells for their folate receptor are overexpressed.Due to the introduction of doxorubicin,DOX @ FA-EuBTC-Pc has a good ability to kill tumor cells.(2)MIL-88B(Fe)is a kind of porous material with trivalent iron as the center metal.The elemental iron is easily available and essential in the human body.In this paper,a kind of iron MOF porous material was synthesized.By the amidation reaction,the phthalocyanine was modified on the material.Trivalent iron can catalyze the decomposition of hydrogen peroxide to generate oxygen,which helps the phthalocyanine photosensitizer to generate more singlet oxygen in the absence of oxygenand and improve anticancer activity.The synthesized materials were characterized by SEM、XRD、IR、XPS、Size and other methods.Then the photodynamic activity of Fe-MOF-Pc nano-systems was studied.The results showed that the Fe-MOF-Pc nanosystem has good photodynamic activity.(3)A novel tetra(4-carboxyphenylalkynyl)phthalocyanine was designed and synthesized.The structures of the compounds were characterized by 1H NMR,13C NMR ang HRMs.Using phthalocyanine as an organic ligand,a novel porous material was constructed,in which the phthalocyanine molecules stand on each other without aggregation.By experiment,an amorphous porous material was prepared and this material was characterized by SEM,XRD and IR.Its ability of generating singlet oxygen at 670 nm laser illumination was tested.The results showed that the material can produce singlet oxygen well under the solid conditions. |
